通过正相硅胶、大孔吸附树脂、MCI树脂、Sephadex LH-20、制备薄层色谱和反相HPLC等多种色谱分离方法相结合,从常用中药党参的水提取物中分离得到16个木脂素类化合物,借助波谱学分析方法鉴定它们的结构分别为:(-)-(7R,7’R,8R,8’S)-4,4-二羟基-3,3’,5,5’,7-五甲氧基-2,7’-环木脂烷(1)、(-)-(7R,8S)-二氢脱氢双松柏基醇4-O-β-吡喃葡萄糖基-(1’’’→2’’)-β-吡喃葡萄糖苷(2)、(-)-(7R,8S)-二氢脱氢双松柏基醇(3)、(+)-(7S,8R)-脱氢二松柏基醇(4)、(+)-蛇菰脂醛素(5)、(+)-去甲氧基松脂素(6)、(+)-松脂素(7)、(+)-表松脂酚(8)、(-)-丁香脂素(9)、(-)-皮树脂醇(10)、(-)-落叶松脂醇(11)、(-)-开环异落叶松脂醇(12)、(-)-对映-异落叶松脂醇(13)、(+)-(7S,8S)-3-甲氧基-3’,7-环氧-8,4’-氧新木脂素-4,9,9’-三醇(14)、(+)-(7S,8R)-3’,4-二羟基-3-甲氧基-8,4’-氧新木脂素(15)和(-)-(7R,8R)-3’,4-二羟基-3-甲氧基-8,4’-氧新木脂素(16)。以上化合物均为首次从党参中分离得到;其中,化合物1为一个新的2,7’-环木脂烷类天然产物,化合物2为一个新的4’,7-环氧-8,3’-新木脂烷类双糖苷。在1×10-5 mol·L-1浓度下,化合物12对半胱氨酸亚铁(Fe~(2+)_Cys)诱导大鼠肝微粒体脂质过氧化的抑制率为(63.4±8.3)%。
Through the chromatographic separation of normal phase silica gel, macroporous adsorption resin, MCI resin, Sephadex LH-20, preparative thin-layer chromatography and reversed-phase HPLC, 16 lignans were isolated from the water extract of common Chinese herb Codonopsis Their structures were identified by spectroscopic analysis as follows: (-) - (7R, 7’R, 8R, 8’S) -4,4-dihydroxy-3,3 ’, 5,5’, 7 (-) - (7R, 8S) -dihydroacenaphthacinol 4-O-β-glucopyranosyl- (1 (3), (+) - (7S, 8R) - (7R, 8S) (+) - dexmethasone (6), (+) - phorbol ester (7), (+) - ) - (-) - linalool (11), (-) - linalool (9), (-) - bercarinol (-) - enantiomer-isomalin (13), (+) - (7S, 8S) -3- methoxy-3 ’, 7-epoxy- Oxyntanosin-4,9,9’-triol (14), (+) - (7S, 8R) -3 ’, 4-dihydroxy-3-methoxy-8,4’-oxo Lignan (15) and (-) - (7R, 8R) -3 ’, 4-dihydroxy-3-methoxy-8,4’-oxylunitignine (16). The above compounds were isolated from Codonopsis for the first time. Among them, compound 1 is a new natural product of 2,7’-cyclopentenoid, and compound 2 is a new 4 ’, 7-epoxy-8,3’ - Neo-lignane bis-glycosides. Under the concentration of 1 × 10-5 mol·L-1, the inhibitory rate of compound 12 on lipid peroxidation in rat liver microsomes induced by Fe2 + Cys was (63.4 ± 8.3) )%.
