目的:研究异黑成熟颗粒对异常黑胆质型慢性阻塞性肺疾病(COPD)大鼠模型炎性反应的影响。方法:用熏烟90d结合气道滴注弹性蛋白酶法建立COPD,在此基础上叠加干寒环境、干寒饮食和足底电刺激建立异常黑胆质型COPD(ASSCOPD)。ASSCOPD于建模后第90天随机分为异黑组和异黑成熟颗粒干预组(给药组),给药组予异黑成熟颗粒灌胃0.54g·200g-1·d-1,共7d。ELISA法检测模型血清和肺泡灌洗液(BALF)中白细胞介素(IL)-8、IL-10、TNF-α。数据用SPSS 17.0统计软件统计分析。结果:(1)血清中炎性因子:与对照组比较:异黑组IL-8含量显著升高(P<0.05)、IL-10、TNF-α含量显著降低(P<0.01,P<0.05),给药组TNF-α含量显著降低(P<0.05)。与模型组比较,异黑组和给药组TNF-α含量显著降低(P<0.05,P<0.01),异黑组IL-8含量显著升高(P<0.05)。与异黑组比较,给药组IL-10含量显著升高(P<0.05)。(2)BALF中炎性因子:与对照组比较,模型组TNF-α含量显著升高(P<0.01),异黑组IL-8、TNF-α含量显著升高(P<0.05,P<0.01),给药组IL-8含量显著升高(P<0.05)。与模型组比较,给药组TNF-α含量显著降低(P<0.05)。结论:异黑成熟颗粒可改善异常黑胆质型COPD大鼠模型的免疫功能,减轻模型炎性反应状态。
Objective: To study the effect of different black mature granules on the inflammatory response in rats with abnormal Savda model of chronic obstructive pulmonary disease (COPD). Methods: COPD was induced by 90 d combined with airway instillation and elastase method. On the basis of this, COPD (ASSCOPD) was established by adding dry and cold environment, dry and cold diet and plantar electrical stimulation. ASSCOPD was randomly divided into different groups (control group) and control group (control group) on the 90th day after model establishment. The rats in the administration group were given intragastric 0.54g · 200g-1 · d-1 intragastrically for 7d . The levels of IL-8, IL-10 and TNF-α in serum and BALF were measured by ELISA. Data using SPSS 17.0 statistical software statistical analysis. Results: (1) Serum inflammatory cytokines: compared with the control group, the content of IL-8 in the different group was significantly increased (P <0.05) and the content of IL-10 and TNF- ), And the content of TNF-α in the treatment group decreased significantly (P <0.05). Compared with the model group, the levels of TNF-αin the different group and the administration group were significantly decreased (P <0.05, P <0.01), while the levels of IL-8 in the different group were significantly increased (P <0.05). Compared with the heterozygous group, the content of IL-10 in the treated group was significantly increased (P <0.05). (2) Inflammatory factor in BALF: Compared with the control group, the content of TNF-α in the model group was significantly increased (P <0.01), the content of IL-8 and TNF-α in the different group was significantly increased (P < 0.01). The content of IL-8 in the treatment group was significantly increased (P <0.05). Compared with the model group, the content of TNF-α in the treatment group was significantly decreased (P <0.05). CONCLUSION: Hematoxylin-eosin mature granules can improve the immune function of model rats with abnormal savda-type COPD and relieve the inflammatory reaction.
