目的:研究青藤碱透过不同实验皮肤的行为。方法:以改进型Franz扩散池为透皮吸收研究实验装置,pH6.8磷酸缓冲液为接收液,采用HPLC法测定试液中青藤碱透过不同实验皮肤的含量。结果:裸鼠的透皮速率常数与人皮较为接近,且重复性较好,其中贮存时间为0、3、7d的裸鼠皮肤的释药速率方程分别为Q=40.34t~(1/2)+19.6、Q=38.01t~(1/2)+25.3、Q=41.55t~(1/2)+24.7。结论:青藤碱透皮吸收研究宜选择裸鼠皮肤,且裸鼠皮肤在-20℃冰箱内贮存7天,对青藤碱的透皮吸收行为不会有明显影响。
Objective: To study the behavior of sinomenine through different experimental skin. Methods: A modified Franz diffusion cell was used as the experimental device for transdermal absorption. pH6.8 phosphate buffer was used as the receiver solution. HPLC was used to determine the content of sinomenine in the test solution through different experimental skin. Results: The transdermal rate constant of nude mice was similar to that of human skin, and the reproducibility was better. The drug release rate equations of nude mice with storage time of 0, 3, and 7 d were Q=40.34t~(1/2), respectively. ) +19.6, Q=38.01t~(1/2)+25.3, Q=41.55t~(1/2)+24.7. Conclusion: The skin of nude mice should be selected for the transdermal absorption of sinomenine, and the skin of nude mice should be stored in the freezer at -20°C for 7 days without significant influence on the transdermal absorption of sinomenine.
