目的:对罗勒多糖进行分离纯化,并进行红外光谱分析。方法:通过水提醇沉、Sevag法除蛋白,再经DEAE离子交换柱层析对多糖进行分离纯化。用红外光谱分析多糖的特征吸收峰。结果:苯酚硫酸法测得罗勒粗多糖含量为28%,考马斯亮蓝测蛋白质含量为10%,用Sevag法除蛋白,纯化率为60%。用DEAE层析柱法纯化,得到中性多糖和酸性多糖,用琼脂糖凝胶电泳法测得结果均为均一多糖。红外光谱分析罗勒多糖具有明显的多糖类物质吸收峰。结论:水提醇沉法提取罗勒多糖提取效率高,可作为罗勒多糖的一种常用提取方法。DEAE离子交换柱对罗勒多糖分离效果较好。
Objective: To isolate and purify basilar polysaccharides and to analyze by infrared spectroscopy. Methods: The polysaccharides were separated and purified by water precipitation, Sevag method and DEAE ion exchange chromatography. Analysis of the characteristic absorption peak of polysaccharides by infrared spectroscopy. Results: The content of basil polysaccharide was 28%, the protein content of Coomassie Brilliant Blue was 10%. The protein was removed by Sevag method and the purification rate was 60%. Purified by DEAE column chromatography to obtain neutral polysaccharides and acidic polysaccharides, agarose gel electrophoresis showed that the results were homogeneous polysaccharides. Infrared spectroscopy basil polysaccharide has obvious absorption peaks of polysaccharides. Conclusion: Extracting basil polysaccharide with water extraction and alcohol precipitation has high extraction efficiency and can be used as a common extraction method of basil polysaccharide. DEAE ion exchange column to basil polysaccharide separation better.
