目的探讨D2-40阳性表达的淋巴管密度(LVD)、C-erbB-2和p53蛋白在大肠癌组织中的表达情况及其生物学意义。方法采用高通量组织芯片技术及免疫组化SP法检测115例大肠腺癌组织(腺癌组)、19例大肠腺瘤组织(腺瘤组)和12例正常大肠黏膜组织(正常组)中D2-40阳性表达的LVD、C-erbB-2和p53蛋白的表达。结果腺癌组中,D2-40阳性表达的LVD、C-erbB-2和p53蛋白的阳性表达率均明显高于腺瘤组和正常组[(24.16±8.43vs.16.78±5.29和12.54±4.62)、(15.65%vs.10.53%和0)和(63.48%vs.21.05%和16.67%)](P<0.05)。D2-40阳性表达的LVD、C-erbB-2和p53蛋白在大肠腺癌中的表达与其分化程度、浸润程度、Dukes分期及淋巴结转移有关。结论 D2-40、C-erbB-2和p53蛋白过表达在大肠癌的发生、发展中起重要作用,可为大肠癌的早期诊断、治疗及预后的判断提供了参考依据。
Objective To investigate the expression of lymphatic vessel density (LVD), the expression of C-erbB-2 and p53 protein in colorectal carcinoma with D2-40 positive expression and its biological significance. Methods High-throughput tissue microarray technique and immunohistochemical SP method were used to detect the expression of VEGF in 115 cases of adenocarcinoma (adenocarcinoma), 19 cases of adenoma (adenoma) and 12 cases of normal colorectal mucosa (normal group) D2-40 positive expression of LVD, C-erbB-2 and p53 protein expression. Results The positive expression rate of LVD, C-erbB-2 and p53 protein in D2-40 positive group was significantly higher than that in adenoma group and normal group [(24.16 ± 8.43 vs.16.78 ± 5.29 and 12.54 ± 4.62 ), (15.65% vs.10.53% and0) and (63.48% vs.21.05% and16.67%, respectively)] (P <0.05). The expression of D2-40 positive LVD, C-erbB-2 and p53 protein in colorectal adenocarcinoma is related to the degree of differentiation, infiltration, Dukes stage and lymph node metastasis. Conclusion Overexpression of D2-40, C-erbB-2 and p53 play an important role in the occurrence and development of colorectal cancer, which may provide a reference for the early diagnosis, treatment and prognosis of colorectal cancer.
