目的:初步研究干细胞相关基因Sox2在膀胱癌的表达及其对膀胱癌细胞体外增殖能力的影响。方法:利用免疫组织化学和免疫荧光法检测Sox2在膀胱癌组织及膀胱癌细胞株T24的表达;将构建成功的pcDNA3.1-SOX2-EGFP质粒转染膀胱癌细胞,采用半定量RT-PCR和Western blot检测Sox2的表达,进一步通过MTT实验初步探讨Sox2对膀胱癌细胞T24体外增殖能力的影响。结果:膀胱癌Sox2高表达率为68.6%;构建的pcDNA3.1-SOX2-EGFP转染膀胱癌细胞T24后能够成功表达;MTT实验显示,Sox2可促进膀胱癌细胞株T24体外增殖(P<0.05)。结论:Sox2在膀胱癌组织及细胞均可检测到表达,并能促进膀胱癌细胞株T24的体外增殖。
Objective: To study the expression of Sox2 in bladder cancer and its effect on the proliferation of bladder cancer cells in vitro. Methods: Immunohistochemistry and immunofluorescence were used to detect the expression of Sox2 in bladder cancer tissue and bladder cancer cell line T24. The constructed pcDNA3.1-SOX2-EGFP plasmid was transfected into bladder cancer cells by semi-quantitative RT-PCR and The expression of Sox2 was detected by Western blot, and the effect of Sox2 on the proliferation of bladder cancer cell T24 in vitro was further investigated by MTT assay. RESULTS: The high expression rate of Sox2 in bladder cancer was 68.6%. The constructed pcDNA3.1-SOX2-EGFP was successfully transfected into bladder cancer T24 cells. MTT assay showed that Sox2 promoted the proliferation of bladder cancer cell line T24 in vitro (P <0.05 ). Conclusion: Sox2 can be detected in bladder cancer tissues and cells, and can promote the proliferation of bladder cancer cell line T24 in vitro.
