目的观察呼吸道合胞病毒(respiratory syncytial virus,RSV)毛细支气管炎(毛支)患儿外周血CD4+CD25+调节性T细胞(Treg)及其分泌细胞因子IL-10、TGF?β的水平,探讨Treg与RSV毛支发病的关系。方法采集31例RSV阳性毛支患儿外周血,采用流式细胞仪检测外周血Treg细胞百分率,ELISA法检测血浆IL-10、TGF?β的水平。27例非RSV感染的普通肺炎患儿作为阳性对照,24例健康儿童为正常对照组。结果 RSV毛支患儿外周血Treg百分率(10.71±1.11)%显著低于肺炎组(12.73±1.35)%及正常对照组(13.24±1.13)%(P<0.01);RSV毛支组血浆IL-10水平(89.83±9.43)ng/L显著低于肺炎组(116.40±14.10)ng/L与正常对照组(124.22±18.38)ng/L(P<0.01);RSV毛支组TGF?β水平(1 019.17±353.32)ng/L显著低于肺炎组(1 979.50±462.30)ng/L与正常对照组(2 139.88±484.75)ng/L(P<0.01)。结论 RSV毛支患儿外周血存在Treg细胞数量及功能的不足,可能是引起RSV毛支发作的重要因素之一。
Objective To investigate the levels of CD4 + CD25 + regulatory T cells (Tregs) and their secreted cytokines IL-10 and TGF-β in peripheral blood of children with respiratory syncytial virus (RSV) bronchiolitis Relationship between Treg and hairy branches of RSV. Methods Totally 31 peripheral blood samples from children with RSV positive branch of hair were collected. The percentage of Treg cells in peripheral blood was detected by flow cytometry. The levels of IL-10 and TGF-β in plasma were detected by ELISA. 27 cases of non-RSV infection in children with common pneumonia as a positive control, 24 healthy children as a normal control group. Results The percentage of Treg in the peripheral blood of children with RSV bronchiolitis was significantly lower than that in the pneumonia group (12.73 ± 1.35)% and the normal control group (13.24 ± 1.13)% (P <0.01) (89.83 ± 9.43) ng / L was significantly lower than that of the pneumonia group (116.40 ± 14.10) ng / L and the normal control group (124.22 ± 18.38) ng / L 1 019.17 ± 353.32) ng / L was significantly lower than that of pneumonia group (1 979.50 ± 462.30) ng / L and the normal control group (2 139.88 ± 484.75) ng / L (P <0.01). Conclusion There is a lack of Treg cell number and function in peripheral blood of children with RSV bronchiolitis, which may be one of the important factors that cause RSV bronchiolitis.
