目的:探讨脑缺血后单核巨噬细胞浸润情况。方法:用免疫组织化学方法检测了30只大鼠大脑中动脉局部脑缺血1 2 h、2 4 h、3d、7d和1 0 d后损伤区ED2 阳性巨噬细胞浸润情况。结果:缺血1 2 h脑实质内可见ED2 阳性巨噬细胞,主要分布于坏死区周围;缺血2 4 h,阳性细胞较1 2 h有所增加,室管膜上有大量的阳性细胞聚集;缺血3d时阳性细胞数达高峰,主要存在于损伤区的周边区。缺血7d、1 0 d时染色中仅偶见阳性细胞的存在。统计学分析大鼠脑缺血1 2 h、2 4 h、3d时ED2 阳性巨噬细胞与正常对照组有非常显著性差异( P<0 .0 1 ) ,缺血3d组与1 2 h组相比差异也有显著意义( P<0 .0 5 )。结论:巨噬细胞参与了缺血性脑损伤的病理过程,主要参与缺血早期的病理损害
Objective: To investigate the infiltration of monocytes after cerebral ischemia. Methods: Immunohistochemistry was used to detect the ED2-positive macrophages infiltration in the damaged area of the middle cerebral artery after focal cerebral ischemia 1, 2, 4, 3d, 7d and 10d in 30 middle cerebral arteries. Results: The ED2-positive macrophages were found in the parenchyma of the brain at 12 h after ischemia, mainly distributed around the necrotic area. At 24 h after ischemia, the number of positive cells increased compared with that at 12 h, and a large number of positive cells gathered on the ependymal membrane The number of positive cells reached the peak at 3d, and mainly existed in the peripheral area of the lesion area. Ischemia 7d, 10d staining only occasional positive cells. Statistical analysis of ED2-positive macrophages at 12 h, 24 h and 3 d after cerebral ischemia in rats showed significant difference from the normal control group (P <0.01) The difference was also significant (P <0.05). CONCLUSION: Macrophages are involved in the pathological process of ischemic brain injury and are mainly involved in pathological damage in the early stage of ischemia
