目的:观察解郁1号方对抑郁模型大鼠海马区白介素-1β(IL-1β)、白介素-2(IL-2)、白介素-6(IL-6)蛋白表达的影响,探讨该方的免疫调节作用。方法:将SD大鼠60只随机分为正常组、模型组、阿米替林组及解郁1号组,每组15只,应用孤养加慢性轻度不可预见性应激方法制备抑郁大鼠模型,采用免疫组化SABC法检测各组大鼠海马区IL-1β、IL-2、IL-6的蛋白表达。结果:与模型组比较,正常组、中西药组大鼠海马CA3区IL-1β、IL-2、IL-6蛋白阳性表达明显,着色较深,阳性表达计数有显著性差异(P<0.01,或P<0.05);中、西药组间相比,IL-1β、IL-2、IL-6蛋白阳性表达计数无明显差异(P>0.05)。结论:解郁1号可能通过提高海马区IL-1β、IL-2、IL-6蛋白表达起免疫调节功能。
Objective: To observe the effect of Jieyu No.1 prescription on the expression of interleukin-1β (IL-1β), interleukin-2 (IL-2) and interleukin-6 (IL-6) in hippocampus of depressive model rats. Immunomodulatory effects. Methods: 60 SD rats were randomly divided into normal group, model group, amitriptyline group and Jieyu No. 1 group. 15 rats in each group were treated with isolation and chronic mild unpredictable stress to prepare depression. In the rat model, the expression of IL-1β, IL-2 and IL-6 in hippocampus of each group was detected by immunohistochemical SABC method. Results: Compared with the model group, the positive expression of IL-1β, IL-2, IL-6 protein in the hippocampal CA3 region of the normal group and the Chinese and Western medicine group was obvious, and the coloration was deeper. The positive expression count had significant difference (P<0.01, Or P<0.05); There was no significant difference in the expression of IL-1β, IL-2, and IL-6 protein between Chinese and Western medicine groups (P>0.05). Conclusion: Jieyu 1 may play an important role in immune regulation by increasing the expression of IL-1β, IL-2 and IL-6 in hippocampus.
