目的:通过观察调肝益气通络方对大鼠间充质干细胞增殖能力的影响,从而评价该方的促增殖能力。方法:腹腔注射“CCl4-橄榄油”混悬液制备Wistar小写大鼠(35只)肝纤维化模型;其中30只灌胃给予调肝益气通络方(低、中、高3个剂量)1个月,制备含药血清;另外5只用于MSC细胞的分离与培养;用含药血清诱导MSC向肝细胞的分化连续7 d;在各检测点进行形态学观察及通过MTT对细胞增殖情况进行评价。结果:各浓度含药血清在加入1~7 d后,其细胞数量均比常规培养组增多,并同用药剂量呈正比,浓度越大细胞数量越多,细胞数随着时间的推移而不断增大。结论:调肝益气通络方对大鼠骨髓里的间充质肝细胞的增殖能力具有显著的促进作用。
OBJECTIVE: To observe the effect of Tiaogan Yiqi Tongluo Recipe on the proliferation of rat mesenchymal stem cells in order to evaluate its promoting ability of proliferation. Methods: Wistar lowercase rats (35 rats) were injected intraperitoneally with “CCl4-olive oil” suspension to prepare hepatic fibrosis model. Thirty rats were given intragastric administration of Tiaogan Yiqi Tongluo Formula (low, middle and high) Dose) for 1 month to prepare serum containing drug; the other 5 cells were used for isolation and culture of MSC cells; MSCs were induced to differentiate into hepatocytes for 7 days with drug-containing serum; morphological observation was performed at each test point and MTT assay Cell proliferation was evaluated. Results: After 1 to 7 days of treatment, the number of cells in each concentration increased more than that in the conventional culture group and was proportional to the dose of drug. The larger the concentration, the more the number of cells and the number of cells increased with the passage of time Big. Conclusion: Tiaogan Yiqi Tongluo Recipe can significantly promote the proliferation of mesenchymal hepatocytes in rat bone marrow.
