基于炎症反应的香椿子总多酚抗大鼠心肌缺血再灌注损伤的机制研究

目的:观察香椿子总多酚对大鼠心肌缺血再灌注急性炎症的影响。方法:50只SD大鼠随机分成假手术组(冠脉下穿线不结扎+0.5%羧甲基纤维素钠)、模型组(冠脉结扎+0.5%羧甲基纤维素钠)、香椿子总多酚低剂量(XD,50 mg.kg-1+冠脉结扎)、中剂量(XZ,100 mg.kg-1+冠脉结扎)、高剂量(XG,200 mg.kg-1+冠脉结扎)组,各组均ig给药。采用左冠状动脉前降支结扎30 min再灌120 min的方法复制大鼠心肌缺血再灌注损伤模型。以ST抬高作为结扎成功的标志,以ST段逐渐回落、梗死性Q波出现作为再灌注成功的标志。再灌结束后行腹主动脉取血,分离血清,测定血清中白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量。取左心室缺血区组织10 mg用RT-PCR测定核因子κB(NF-κB)p65 mRNA的表达水平,其余心肌组织做病理切片,并在光镜下观察左心室心肌组织的形态学变化。结果:模型组大鼠血清IL-6含量为(638.88±188.94)ng.L-1,XD,XZ,XG组与模型组相比,血清IL-6含量显著下降,分别为(491.58±142.59),(306.85±80.60),(246.11±71.64)ng.L-1。模型组大鼠血清TNF-α含量为(216.23±58.67)ng.L-1,XD,XZ,XG组大鼠血清TNF-α水平与之相比显著降低,分别为(148.24±36.30),(157.62±52.06),(144.10±36.64)ng.L-1。模型组大鼠心肌组织中NF-κBp65/β-actin为0.61±0.05,香椿子总多酚给药组大鼠NF-κB p65/β-actin与之相比显著降低,分别为0.53±0.08,0.45±0.13,0.38±0.11。香椿子总多酚组与模型组相比,大鼠心肌细胞的形态学损伤较轻。结论:香椿子总多酚能够减轻大鼠心肌缺血再灌注急性炎症,对其产生一定的保护作用。 Objective: To observe the effect of total polyphenols of Toona sinensis on acute inflammation of myocardial ischemia-reperfusion in rats. Methods: Fifty Sprague-Dawley rats were randomly divided into sham operation group (non-ligated coronary artery + 0.5% sodium carboxymethyl cellulose), model group (coronary artery ligation + 0.5% sodium carboxymethyl cellulose) Low dose polyphenols (XD, 50 mg.kg -1 + coronary ligation), middle dose (XZ, 100 mg.kg -1 + coronary ligation), high dose (XG 200 mg.kg -1 + coronary artery Ligation) group, each group were ig administration. The left anterior descending coronary artery was ligated for 30 min and then reperfused for 120 min to duplicate the model of myocardial ischemia-reperfusion injury in rats. To ST elevation as a sign of successful ligation, ST segment gradually decreased, infarction Q wave appeared as a sign of reperfusion success. After reperfusion, blood was taken from the abdominal aorta and the serum was separated to determine the levels of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in serum. The expression of nuclear factor kappaB (NF-κB) p65 mRNA was determined by RT-PCR in 10 mg of left ventricular ischemic area. The remaining myocardial tissue was examined by pathology. The morphological changes of left ventricular myocardium were observed under light microscope. Results: The level of IL-6 in model group was (638.88 ± 188.94) ng.L-1, the level of IL-6 in XD, XZ and XG groups was significantly lower than that in model group (491.58 ± 142.59, , (306.85 ± 80.60), (246.11 ± 71.64) ng.L-1. The level of serum TNF-αin the model group was (216.23 ± 58.67) ng.L-1, the levels of TNF-α in XD, XZ and XG groups were significantly lower than that in the model group (148.24 ± 36.30, 157.62 ± 52.06), (144.10 ± 36.64) ng.L-1. The NF-κB p65 / β-actin in the myocardium of model group was 0.61 ± 0.05, and the level of NF-κB p65 / β-actin in the total polyphenol treatment group was significantly lower than that in the model group (0.53 ± 0.08, 0.45 ± 0.13 and 0.38 ± 0.11. Toon seed total polyphenol group compared with the model group, the morphological damage of rat cardiomyocytes lighter. CONCLUSION: Total polyphenols from Toona sinensis can relieve acute inflammation of myocardial ischemia-reperfusion injury in rats, and have a certain protective effect on it.
