目的探讨苏氨酸双铜配合物对糖尿病小鼠血糖、血脂的影响以及对心肾血管的保护作用。方法采用四氧嘧啶腹腔注射制作小鼠糖尿病模型。灌胃给与糖尿病模型小鼠低、中、高(0.002%,0.02%,0.1%)三种剂量的苏氨酸双铜配合物,测定给药 0,30及45天的空腹血糖,45天处死动物取血及心、肾组织,测定糖化血红蛋白、血脂及观察不同剂量药物对糖尿病小鼠的心、肾病理改变的影响。结果与蒸馏水灌胃对照组比较:苏氨酸双铜配合物中剂量组降血糖作用最显著(P<0.01),低剂量组降血糖作用次之(P<0.05);苏氨酸双铜配合物低、中、高剂量组胆固醇(TC)水平明显降低(P<0.05),高密度脂蛋白(HDL)水平明显升高(P<0.05);中剂量组低密度脂蛋白(LDL)水平明显降低(P<0.05);中剂量组心肌纤维排列整齐,细胞核和细胞浆分布均匀,肾小球结构正常,细胞大小分布均匀,染色正常。结论苏氨酸双铜配合物具有显著的降血糖、调节血脂紊乱的作用,并对糖尿病小鼠的心肾组织起到明显的保护作用,其中以中剂量组(0.02%)效果最佳。
Objective To investigate the effects of threonine bis-copper complex on blood glucose and blood lipid in diabetic mice and its protective effects on the heart and renal blood vessels. Methods Alloxan were intraperitoneally injected into diabetic mice model. Low, medium and high (0.002%, 0.02%, 0.1%) threonine bis-copper complexes were administered to diabetic mice by intragastric administration. Fasting plasma glucose was measured on days 0, 30 and 45 after administration for 45 days Animals were sacrificed to take blood and heart, kidney, determination of glycosylated hemoglobin, blood lipids and observed different doses of drugs on diabetic mice heart and kidney pathological changes. Results Compared with the distilled water control group, the effect of reducing blood sugar was the most significant (P <0.01) and the second one was the second (P <0.05), the double dose of threonine double copper complex The level of cholesterol (TC) in low, medium and high dose group was significantly lower (P <0.05) and the level of high density lipoprotein (HDL) was significantly increased (P <0.05) (P <0.05). The myocardial fibers of middle-dose group were arranged neatly with uniform distribution of nucleus and cytoplasm, normal glomerular structure, uniform cell size distribution and normal staining. Conclusions Threonine bis-copper complex has significant hypoglycemic effect and regulates dyslipidemia, and has obvious protective effect on heart and kidney of diabetic mice. The medium-dose group (0.02%) has the best effect.
