目的观察热休克蛋白gp96(HSPgp96)在人骨肉瘤组织中的表达。方法采用免疫组织化学SP法检测45例骨肉瘤组织,其中骨母细胞型24例,软骨母细胞型20例,纤维母细胞型1例,以及5例正常骨组织中HSPgp96的表达情况。结果HSPgp96在正常骨组织与骨肉瘤组织中的表达率分别为20%(1/5)及100%(45/45),平均阳性细胞率分别为0.48%和97.23%,两者差异有统计学意义(P<0.01),在骨母细胞型、软骨母细胞型与纤维母细胞型中阳性表达率之间差异无统计学意义。HSPgp96在骨肉瘤中的表达与病理分级明显相关(P<0.05),与临床分期无关(P>0.05)。结论HSPgp96在人骨肉瘤组织中高表达,可作为判断恶性程度的重要指标以及肿瘤免疫的重要靶位。
Objective To observe the expression of heat shock protein gp96 (HSPgp96) in human osteosarcoma. Methods Immunohistochemical SP method was used to detect the expression of HSPgp96 in 45 cases of osteosarcoma, including 24 cases of osteoblast type, 20 cases of chondroblast cell type, 1 fibroblast type and 5 cases of normal bone tissue. Results The positive rates of HSPgp96 in normal and osteosarcoma tissues were 20% (1/5) and 100% (45/45), respectively. The average positive rates of HSPgp96 were 0.48% and 97.23% (P <0.01). There was no significant difference in the positive rates of osteoblastic, chondroblastic and fibroblast types between osteoblasts and fibroblasts. The expression of HSPgp96 in osteosarcoma was significantly correlated with pathological grade (P <0.05), but not with clinical stage (P> 0.05). Conclusion HSPgp96 is overexpressed in human osteosarcoma tissue and may serve as an important indicator of malignancy and an important target of tumor immunity.
