为探讨神经肽 Y 对血管壁平滑肌细胞凋亡相关基因表达的诱导作用,以体外培养血管平滑肌细胞模型为研究对象,运用免疫荧光组织化学技术和激光共聚焦显微镜定量研究神经肽 Y 对血管平滑肌细胞凋亡相关基因bcl- 2 、bax 和fas 表达的影响,以阐明神经肽 Y 参与高血压和动脉粥样硬化等心血管疾病发生的分子机制。结果发现,神经肽 Y 浓度为10 - 6 mol/ L 时,血管平滑肌细胞凋亡相关基因bcl- 2 和bax 表达的平均荧光值分别为1834 .25 ±138 .55 和1870 .82 ±162 .43 ,明显高于对照组的1682 .87 ±94 .47 和1688 .04 ±110 .91( P< 0 .01) ;当神经肽 Y 浓度为10 - 7 mol/ L 时,bcl- 2 和bax 的表达降低( P< 0 .05) ;而神经肽 Y 浓度为10 - 8 mol/ L 时,bcl- 2 和bax 的表达已接近对照组的表达水平( P> 0 .05) 。但fas 仅在神经肽 Y 浓度为10 - 6 mol/ L时,其平均荧光值为1987 .68 ±185 .39 ,与对照组的1882 .38 ±160 .84 相比明显增高( P< 0 .05) 。结果提示,神经肽 Y可诱导体外培养的
