[目的]观察裙带菜多糖(PUP)对食管癌细胞TE-13的生长抑制作用,探讨其诱导细胞周期阻滞和细胞凋亡的机制。[方法]以食管癌细胞TE-13作为研究对象,MTT法检测PUP对其增殖的抑制作用;在电子透镜下观察PUP对TE-13超微凋亡结构的影响;流式细胞技术检测其周期分布和凋亡率;RT-PCR法检测PUP对TE-13细胞中凋亡相关基因Survivin表达的影响。[结果]PUP对食管癌细胞TE-13有显著增殖抑制效应,作用时间越长,在一定范围内的PUP浓度越大,抑制效应越强。PUP作用48h时半数抑制浓度(IC50)为52.17μg/ml。25、50、100μg/ml的PUP作用48h后,TE-13均发生凋亡形态的变化;100μg/ml PUP处理72h后,凋亡率为52.19%;周期分布显示G0/G1期细胞增多,S期细胞明显减少。RT-PCR结果显示PUP能降低食管癌细胞中Survivin mRNA的表达。[结论]PUP在体外显著抑制食管癌细胞TE-13的增殖,其机制与PUP诱导食管癌周期阻滞和下调Survivin mRNA的表达有关。
[Objective] To observe the inhibitory effect of Undaria pinnatifida polysaccharides (PUP) on the growth of esophageal cancer cell line TE-13 and to explore its mechanism of inducing cell cycle arrest and apoptosis. [Method] The esophageal cancer cell line TE-13 was used as the research object. The inhibitory effect of PUP on the proliferation was detected by MTT assay. The effect of PUP on the apoptosis of TE-13 cells was observed under the electron lens. The cell cycle was detected by flow cytometry Distribution and apoptosis rate. The effect of PUP on Survivin expression in TE-13 cells was detected by RT-PCR. [Result] PUP had a significant inhibitory effect on esophageal cancer cells TE-13. The longer the effect, the greater the concentration of PUP in a certain range, and the stronger the inhibitory effect. The half inhibitory concentration (IC50) at 48h was 52.17μg / ml. After treated with 25, 50 and 100μg / ml of PUP for 48h, the morphological changes of TE-13 all occurred. After the treatment with 100μg / ml PUP for 72h, the apoptosis rate was 52.19%. The cell cycle distribution showed that G0 / Period of cells was significantly reduced. The results of RT-PCR showed that PUP can reduce the expression of Survivin mRNA in esophageal cancer cells. [Conclusion] PUP significantly inhibits the proliferation of esophageal cancer cells TE-13 in vitro. The mechanism of PUP is related to the cycle arrest of PUP-induced esophageal cancer and the down-regulation of Survivin mRNA expression.
