目的观察大肠癌组织及正常结直肠组织标本中外周型苯二氮受体(PBR)、p27Kip1、Cyclin E的表达,探讨大肠癌可能的临床预后影响因素。方法收集85例手术治疗大肠癌组织及15例正常结直肠组织标本,进行免疫组化染色,观察p27Kip1、Cyclin E蛋白的表达;收集40例大肠癌患者的癌组织及10例正常结直肠组织新鲜冰冻标本,蛋白质印迹分析检测PBR、p27Kip1和Cyclin E蛋白表达,RT-PCR检测PBR、p27Kip1、Cyclin E mRNA的表达。结果大肠癌组织中PBR、Cyclin E高表达,p27Kip1低表达,且Cyclin E与p27Kip1蛋白表达负相关;Cyclin E阴性和p27Kip1阳性患者的5年生存率明显增加;p27Kip1和Cyclin E的表达进入Cox风险比例模型。结论大肠癌组织PBR、p27Kip1、Cyclin E异常表达。
Objective To observe the expressions of peripheral blood benzodiazepine receptor (PBR), p27Kip1 and Cyclin E in colorectal cancer tissues and normal colorectal tissues and to explore the possible prognostic factors of colorectal cancer. Methods Totally 85 cases of colorectal cancer and 15 normal colorectal tissues were collected for immunohistochemical staining to observe the expression of p27Kip1 and Cyclin E protein. Forty cases of colorectal cancer and 10 cases of normal colorectal tissue were collected for fresh The expression of PBR, p27Kip1 and Cyclin E protein were detected by Western blot analysis and RT-PCR. The expression of PBR, p27Kip1 and Cyclin E mRNA were detected by RT-PCR. Results The expressions of PBR and Cyclin E were high, p27Kip1 was low, and Cyclin E was negatively correlated with p27Kip1 in colorectal carcinoma. The 5-year survival rates of Cyclin E-negative and p27Kip1-positive patients were significantly increased. The expressions of p27Kip1 and Cyclin E were associated with Cox risk Proportional model. Conclusions Abnormal expression of PBR, p27Kip1 and Cyclin E in colorectal cancer tissues.
