为进一步探究绵羊肺炎支原体P108蛋白的结构及其免疫原性,在进行生物信息学分析的基础上,对编码P108的部分基因片段进行了PCR扩增、克隆和原核表达,并采用Western-blot方法对其免疫原性进行了分析。结果显示,p108基因与猪肺炎支原体黏附相关基因mhp385的核苷酸同源性较高,扩增的p108部分基因在大肠杆菌BL21(DE3)中成功获得表达,重组蛋白以包涵体的形式存在;重组蛋白可以与山羊抗绵羊肺炎支原体血清发生结合反应,表明P108蛋白是免疫原之一。本研究结果表明P108蛋白可以诱导机体产生免疫应答,是建立绵羊肺炎支原体感染血清学诊断技术的潜在抗原。
In order to further explore the structure and immunogenicity of Mycoplasma pneumoniae P108 protein, the partial P108 gene fragment was amplified by PCR, cloned and prokaryotic expressed on the basis of bioinformatics analysis and Western-blot method Its immunogenicity was analyzed. The results showed that the nucleotide sequence of p108 gene was highly homologous to Mycoplasma hyopneumoniae adhesion-related gene mhp385. The amplified part of p108 gene was successfully expressed in E. coli BL21 (DE3), and the recombinant protein existed in the form of inclusion body. The recombinant protein can bind to the goat anti-Mycoplasma hominis serum, indicating that the P108 protein is one of the immunogens. The results of this study indicate that P108 protein can induce the body to produce an immune response and is a potential antigen for establishing a serological diagnostic technique for Mycoplasma pneumoniae infection.
