目的 探讨紫杉醇对Lewis肺癌生长和转移的抑制作用。方法 将Lewis肺癌细胞接种于C5 7BL/6小鼠皮下 ,腹腔注射紫杉醇 1 0mg/kg ,连续 1 0d。处理动物后测量治疗组及对照组皮下瘤重、肺转移灶计数 ,应用流式细胞术测定肿瘤细胞凋亡率 ,并进行光镜和电镜观察。结果 对照组和治疗组皮下瘤重分别为 (2 .833± 0 .0 74)g和 (0 .5 88± 0 .0 40 )g(P <0 .0 1 ) ,肺转移灶计数分别为 (5 .5 0 0± 0 .92 6 )个和(1 .1 6 7± 0 .75 3)个 (P <0 .0 1 ) ,肿瘤细胞凋亡率分别为 9.0 0 2 %和 2 5 .772 % (P <0 .0 1 )。电镜观察紫杉醇组可见典型的凋亡细胞和凋亡小体。结论 紫杉醇可明显抑制小鼠Lewis肺癌的生长和转移 ,并可诱导肿瘤细胞凋亡
Objective To investigate the inhibitory effect of paclitaxel on the growth and metastasis of Lewis lung cancer. Methods Lewis lung cancer cells were inoculated subcutaneously into C57BL/6 mice and intraperitoneal injection of paclitaxel 10 mg/kg for 10 days. After treatment of animals, subcutaneous tumor weights and lung metastases were counted in the treatment group and the control group. Flow cytometry was used to determine the apoptosis rate of tumor cells, and light and electron microscopy were performed. Results The subcutaneous tumor weights of the control group and the treatment group were (2.833±0.074)g and (0.58±0.040)g, respectively (P <0. 01). The lung metastases counts were (5.5 0 ± 0 .92 6) and (1.167 ± 0.753) (P <0. 01), with apoptotic rates of 9.0 0 2% and 2 5, respectively. .772% (P < 0.01). Electron microscopy showed typical apoptotic cells and apoptotic bodies in the paclitaxel group. Conclusion Paclitaxel can significantly inhibit the growth and metastasis of Lewis lung cancer in mice and induce tumor cell apoptosis.
