目的:对千金子中千金子素L1(化合物Ⅰ)、千金子素L2(化合物Ⅱ)和千金子素L3(化合物Ⅲ)3种千金二萜醇酯进行含量测定。方法:采用反相高效液相色谱法,使用Prevail-C18柱(250mm×4.6mm,5μm),流动相为甲醇-水(75∶25),流速1.0mL·min-1,检测波长280nm,柱温25℃。结果:化合物Ⅰ、Ⅱ、Ⅲ进样量均在0.1~4.0μg范围内有良好的线性关系;相关系数r分别为0.9998,0.9999,0.9996;平均回收率(n=6)分别为97.9%(RSD为1.5%),100.9%(RSD为1.5%),99.6%(RSD为1.2%)。结论:本方法能够准确测定千金子中化合物Ⅰ、Ⅱ、Ⅲ3种千金二萜醇酯的含量。
OBJECTIVE: To determine the contents of three kinds of chalcone esters of Lutein L1 (Compound I), Lutein L2 (Compound II) and Lutein L3 (Compound III) in Semen Trigonella. Methods: The reversed-phase high performance liquid chromatography was used on a Prevail-C18 column (250mm×4.6mm, 5μm). The mobile phase was methanol-water (75∶25) with a flow rate of 1.0mL·min-1. The detection wavelength was 280nm. Temperature 25 °C. RESULTS: Compounds I, II, and III had good linearity in the range of 0.1-4.0 μg; the correlation coefficients r were 0.9998, 0.9999, and 0.9996, respectively; the average recoveries (n=6) were 97.9% (RSD, respectively). It was 1.5%), 100.9% (RSD 1.5%), and 99.6% (RSD 1.2%). Conclusion: This method can accurately determine the contents of compounds I, II, III, and three kinds of diterpene alcohol esters.
