目的:考察大鼠灌服芎汤超临界提取物后,其有效成分藁本内酯、正丁基苯酞和洋川芎内酯A在体内的排泄动力学规律。方法:将超临界部位对SD大鼠灌胃给药后,按不同时间段收集大鼠的尿液和粪样,对以上3种成分进行HPLC测定,绘制其尿和粪样排泄规律曲线,考察排泄特征。结果:藁本内酯0~24 h尿排泄率逐渐增高,24 h后逐渐降低,在粪样中36~48 h排泄率最高,此后逐渐降低;正丁基苯酞在尿液中12~48 h排泄率较高;洋川芎内酯A在粪样中12~48 h排泄率较高。结论:尿液中藁本内酯和正丁基苯酞,粪样中藁本内酯和洋川芎内酯A累积排泄量-时间曲线、平均排泄率-时间曲线比较一致,反映出共性的排泄特征,可为芎汤超临界提取物的整体排泄特征提供参考。
OBJECTIVE: To investigate the excretion kinetics of ligustilide, n-butylphthalide and yangchongxionglactone A in rats after oral administration of the supercritical extract of Xiongxiong decoction in rats. Methods: After oral administration of supercritical fluid to SD rats, the urine and feces samples of rats were collected at different time points. The above three components were determined by HPLC, and their urinary and excretion patterns were drawn. Excretion characteristics. Results: The urinary excretion rate of ligustilide gradually increased from 0 to 24 hours and decreased gradually after 24 hours. The excretion rate of excretion was the highest from 36 to 48 hours in fecal samples, and then decreased gradually. The content of n-butylphthalide in urine ranged from 12 to 48 h excretion rate higher; Yang Chuanxiong lactone A in fecal sample 12 to 48 h excretion rate higher. CONCLUSION: The cumulative excretion-time curves and mean excretion rate-time curves of ligustilide and n-butylphthalide in urine, fecal samples of ligustilide and yangchongxiong lactone A are consistent, which reflect the common excretion characteristics, It can provide a reference for the overall excretion characteristics of Xiong Gui Tang supercritical extract.
