目的:研究胃癌病人血清mi R-338-3p的表达及其对胃癌细胞生物学行为的影响。方法:提取40例胃癌病人及37例健康志愿者血清总mi RNA,采用逆转录实时荧光定量聚合酶链反应检测mi R-338-3p的表达。基于生物信息学预测其功能,采用细胞转染、侵袭、增殖实验及流式细胞术、Western印迹,研究其对胃癌细胞生物学行为的影响。结果:胃癌病人血清mi R-338-3p表达显著低于健康志愿者(P<0.001)。体外实验证明,转染mi R-338-3p后可明显抑制胃癌细胞株SGC7901和BGC823增殖(P<0.01)以及细胞侵袭能力,并促进其凋亡。同时抑制基质金属蛋白酶(matrix metalloproteinase,MMP)2蛋白的表达(P<0.05)。结论:胃癌病人血清mi R-338-3p表达明显降低。mi R-338-3p具有抑制胃癌细胞增殖和侵袭、促进凋亡的作用。可能通过下调MMP2表达抑制胃癌细胞。
Objective: To study the expression of mi R-338-3p in gastric cancer patients and its effect on the biological behavior of gastric cancer cells. Methods: Total mi RNA was extracted from 40 patients with gastric cancer and 37 healthy volunteers. The expression of mi R-338-3p was detected by reverse transcription real-time polymerase chain reaction. Based on bioinformatics, we predicted its function by cell transfection, invasion and proliferation assay, flow cytometry and Western blotting, and studied its effect on the biological behavior of gastric cancer cells. Results: The expression of mi R-338-3p in gastric cancer patients was significantly lower than that in healthy volunteers (P <0.001). In vitro experiments showed that mi R-338-3p transfected gastric cancer cells SGC7901 and BGC823 significantly inhibited the proliferation (P <0.01) as well as cell invasiveness and apoptosis. While inhibiting the expression of matrix metalloproteinase (MMP) 2 protein (P <0.05). Conclusion: The expression of mi R-338-3p in patients with gastric cancer is significantly lower. mi R-338-3p can inhibit the proliferation and invasion of gastric cancer cells and promote apoptosis. It may inhibit gastric cancer cells by down-regulating MMP2 expression.
