目的:研究X线照射对结直肠癌HCT116细胞中肿瘤转移抑制基因Kiss-1表达的影响,分析Kiss-1基因表达变化与结直肠癌侵袭、迁移潜能的关系。方法结直肠癌HCT116细胞经4Gy、8Gy剂量X线照射后24h、48h,应用免疫细胞化学法检测Kiss-1基因表达水平的变化,Transwel 小室侵袭和迁移实验比较X线照射后HCT116细胞侵袭、迁移潜能的改变。结果与对照组癌细胞相比,X照射后24h、48h的4Gy、8Gy剂量组癌细胞中Kiss-1基因表达水平均明显上调(P<0.05),癌细胞的侵袭、迁移能力均明显降低(P<0.05)。结论 X线照射上调结直肠癌细胞中转移抑制基因Kiss-1的表达,残存癌细胞中Kiss-1基因的表达增强有可能抑制了癌细胞的转移潜能。“,”Objective To investigate the change of metastasis suppressor gene Kiss-1 protein expression level in human colorectal cancer cells and organization irradiated by X-ray, and analysis its influence for the ability of migration and invasion of colorectal carcinoma. Methods HCT116 was irradiated at different does (4, 8Gy), after 24 and 48h, the cellimmunohistochemistry was used to find out the effect of X-ray for the Kiss-1 protein expression; the Transwel invasion model was established by Transwel chamber of 8 um diameter and ECM as the basement membrane, was used to observe the invasion of HCT116; the Transwel migration model was established by the Transwel chamber of 8um diameter, was used to observed the migration of HCT116. Result: Compare with the control group, 24h and 48h after radiation, Kiss-1 protein expression was increased for 4Gy group and 8Gy group cells (P<0.05). In the migration and invasion assay, the migration and invasion of HCT116 cells were significantly decreased after irradiation including 4Gy, 8Gy group (P<0.05). Conclusion X-ray Radiation could increase expression of Kiss-1 protein in colorectal cancer cell, and the ability of metastasis were decreased for HCT116 cells which were survival after irradiated.
