目的观察复方丹参滴丸及其组成药物丹参、三七对豚鼠离体心脏心电图的影响,研究其心脏毒性作用。方法应用Langendorff离体豚鼠心脏灌流装置,将复方丹参滴丸浸膏和丹参浸膏(给药质量浓度均以丹参素计)、三七浸膏(给药质量浓度以人参皂苷Rg1计)均按3、0.3、0.1、0.03μg/mL灌注,观察离体豚鼠心脏心电图的变化。结果复方丹参滴丸浸膏和丹参浸膏对豚鼠离体心脏心电图无明显影响,三七浸膏在低质量浓度(0.3、0.1、0.03μg/mL)对豚鼠离体心脏心电图没有影响,但在3μg/mL时可轻微延长PR、RR间期,减慢心率,与对照相比差异显著(P<0.05)。3种药物对校正后的QT间期(QTc)及T波峰到T波结束间期(Tp-e)均无显著影响。结论复方丹参滴丸及其组成药物丹参、三七在实验质量浓度范围内不诱发心室肌细胞复极化延长及严重心律失常。
Objective To observe the effects of compound Danshen dripping pills and its constituent drugs of Salvia miltiorrhiza and Sanchi on the isolated cardiogram of guinea pigs and to study its cardiotoxic effect. Methods Langendorff guinea pig heart perfusion device was used. The compound Danshen dripping pills and Salvia miltiorrhiza extract (the dosage of Danshensu were counted), the notoginseng extract (the dosage of ginsenoside Rg1) 3,0.3,0.1,0.03μg / mL perfusion, observed guinea pig heart electrocardiogram changes. Results Compound Danshen Diwan extract and Salvia miltiorrhiza extract had no significant effect on isolated heart electrocardiogram in guinea pigs. The low concentration of Sanqi (0.3,0.1,0.03μg / mL) had no effect on isolated heart electrocardiogram in guinea pigs. However, 3μg / mL can slightly prolong the PR, RR interval, heart rate, compared with the control significantly (P <0.05). The three drugs had no significant effect on the corrected QTc and T wave to Tp-e. Conclusion Compound Danshen Dripping Pill and its components drug Danshen and Sanqi do not induce prolonged repolarization of ventricular myocytes and severe arrhythmia in the experimental mass concentration range.
