目的建立苦参汤有效部位中4种黄酮类成分黄芩苷、汉黄芩苷、黄芩素和汉黄芩素的HPLC含量测定方法。方法采用Phenomenex Luna C18柱(4.6 mm×250 mm,5μm),甲醇-磷酸水溶液[磷酸-水(2∶530)](45∶55)为流动相,流速1 mL/m in,检测波长280 nm。结果黄芩苷平均回收率为99.28%,RSD=1.92%(n=5),汉黄芩苷平均回收率为98.48%,RSD=1.53%(n=5),黄芩素平均回收率为99.51%,RSD=1.81%(n=5),汉黄芩素平均回收率为97.43%,RSD=2.14%(n=5)。结论3批样品测定结果表明,该法简便、准确,可用于苦参汤有效部位中黄芩苷等4种黄酮类成分含量测定。
Objective To establish a HPLC method for determination of four flavonoids baicalin, wogonin, baicalein and wogonin in the effective fraction of Kushen Tang. Method Phenomenex Luna C18 column (4.6 mm×250 mm, 5 μm), methanol-phosphoric acid aqueous solution [phosphoric acid-water (2:530)] (45:55) as mobile phase, flow rate 1 mL/min, detection wavelength 280 nm . Results The average recovery rate of baicalin was 99.28%, RSD was 1.92% (n=5), the average recovery rate of wogonin was 98.48%, RSD was 1.53% (n=5), and the average recovery rate of baicalein was 99.51%. =1.81% (n=5), the average recovery of wogonin was 97.43%, and RSD was 2.14% (n=5). Conclusion The determination results of three batches of samples showed that the method is simple and accurate, and can be used for the determination of four flavonoids such as baicalin in the effective part of Kushen Tang.
