目的 研究雷公藤红素对狼疮鼠肾小球Ⅰ型胶原 (ColⅠ )和Ⅳ型胶原 (ColⅣ )的影响。方法 在不同发病时间、用不同剂量的雷公藤红素对BWF1小鼠进行腹腔注射。每 2周检测小鼠 2 4h尿蛋白量和血清抗dsDNA抗体水平 ;2 0周后检测其肾组织ColⅠ、ColⅣ、基质金属蛋白酶 2(MMP 2 )及其组织抑制剂 2 (TIMP 2 )的表达 ;反转录 巢式PCR法检测小鼠肾局部转化生长因子(Transforminggrowthfactor,TGF) β1mRNA的表达。结果 ①雷公藤红素抑制小鼠蛋白尿及血清抗dsD NA抗体水平 ,增加肾局部MMP 2而减少ColⅠ、ColⅣ、TIMP 2及TGF β1的表达 ,改善小鼠肾组织学病变。②蛋白尿发生前使用雷公藤红素较蛋白尿发生后使用疗效更显著。③蛋白尿发生前每周使用雷公藤红素 3mg/kg和 6mg/kg的疗效无统计学差异。结论 雷公藤红素对该狼疮模型的肾小球硬化具有明确保护作用。其降低肾脏ColⅠ、ColⅣ的沉积可能是通过增加小鼠肾局部MMP 2、抑制TGF β1及TIMP 2的表达而实现的
Objective To study the effects of tripterine on glomerular collagen Ⅰ (ColⅠ) and collagen Ⅳ (Col Ⅳ) in the lupus. Methods BWF1 mice were injected intraperitoneally with different doses of tripterine at different time of onset. The levels of urinary protein and serum anti-dsDNA antibody were measured every 24 weeks. The expression of ColⅠ, ColⅣ, MMP 2 and tissue inhibitor of metalloproteinase 2 (TIMP 2) Reverse transcriptase nested PCR was used to detect the expression of transforming growth factor β1 mRNA in mouse kidney. Results Tripton inhibited mouse proteinuria and serum anti-dsD NA antibody levels, increased local MMP 2 and decreased ColⅠ, ColⅣ, TIMP 2 and TGFβ1 expression, and improved renal histological lesions. ② the use of triptolide before proteinuria occurred more significantly after the use of proteinuria. ③ proteinuria week before the use of triptolide 3mg / kg and 6mg / kg of no significant difference in the efficacy. Conclusion Tripterine has a clear protective effect on glomerulosclerosis in this lupus model. Decreasing the deposition of ColⅠand ColⅣ in the kidney may be achieved by increasing the local MMP 2 and inhibiting the expression of TGFβ1 and TIMP 2 in the kidney of mice
