目的:建立复方维A酸脂质体中蒿酮的含量和包封率测定的高效液相色谱(High performance liquid chromatography,HPLC)法。方法:色谱柱:迪马Kromasil C18柱(250 mm×4.6 mm,5μm);流动相:乙腈-水(75∶25);检测波长:242 nm;柱温:30℃;流速:0.8 ml/min;进样量:10μl。结果:蒿酮在25.16~251.60μg/ml范围内具有良好的线性关系(r=0.999 9),平均回收率为99.53%,RSD为0.72%。结论:该方法简便快速、准确,具有良好的重复性和回收率,可作为该制剂中蒿酮的定量分析方法。
OBJECTIVE: To establish a high performance liquid chromatography (HPLC) method for the determination of the content of artesunate and entrapment efficiency in the compound retinoic acid liposomes. Method: Column: Dima Kromasil C18 column (250 mm × 4.6 mm, 5 μm); mobile phase: acetonitrile-water (75:25); detection wavelength: 242 nm; ; Injection volume: 10μl. Results: Artemisia ketone had a good linear relationship (r = 0.999 9) in the range of 25.16 ~ 251.60μg / ml with the average recovery of 99.53% and RSD 0.72%. Conclusion: The method is simple, rapid, accurate, and has good repeatability and recovery rate. It can be used as a quantitative analysis method of artemisinin in the preparation.
