[目的]观察千金文武汤对胰岛素抵抗模型大鼠瘦素表达水平的影响.[方法]用高脂饲料喂养建立胰岛素抵抗大鼠模型.给予千金文武汤干预治疗,以罗格列酮作为阳性对照组,采用高胰岛素-正常血糖钳夹实验检测大鼠葡萄糖输注率(GIR).取材附睾旁脂肪组织进行瘦素PCR,以观察千金文武汤对胰岛素抵抗大鼠模型的敏感性和脂代谢的影响.[结果]经钳夹鉴定胰岛素抵抗模型建立,千金文武汤组与罗格列酮组的GIR较模型对照组显著增高(P<0.05,P<0.01),瘦素PCR扩增后积分光密度表达、mRNA表达水平间差异均有统计学意义(P<0.05,P<0.01).[结论]千金文武汤提高胰岛素抵抗大鼠模型的胰岛素敏感性,改善瘦素水平的高表达.与罗格列酮组相比较在脂代谢和瘦素调节上存在差异,与千金文武汤的作用靶点在调节脂代谢和脂肪因子有关联.
[Objective] To observe the effect of Qianjin Wenwu decoction on the expression of leptin in insulin resistance model rats. [Methods] The model of insulin resistance was established by feeding with high-fat diet and treated with Qianjinwenwu Decoction. Rosiglitazone was used as positive control Group, the insulin infusion rate (GIR) of rats was measured by high glucose-insulin clamp and the leptin PCR was performed on the adipose tissue of the epididymis to observe the sensitivity and the lipid metabolism of Qianjinwenwu Tang to insulin resistance model rats (P <0.05, P <0.01). After the leptin PCR amplification, the integral light intensity was significantly higher than that of the model control group (P <0.05, P <0.01). [Conclusion] Qianjinwenwu decoction can improve the insulin sensitivity of insulin resistance rat model and improve the high expression of leptin level.Compared with Luo Glitazone compared lipid metabolism and leptin in the regulation of the differences, and Qian Jin Wen Wu Tang’s role in the regulation of lipid metabolism and adipokines are related.
