目的人白血病抑制因子(LIF)基囚在COS-7细胞中进行瞬时表达,检测表达产物对HL-60白血病细胞的作用。方法用脂质体转染法将pcDNA3.0-LIF真核表达质粒导入COS-7细胞,通过形态学观察、MTT、FCM法研究COS-7细胞表达产物对HL-60细胞的影响。结果人LIF基因可在COS-7细胞中进行有效表达,表达产物对HL-60白血病细胞具有抑制生长和诱导凋亡的作用。结论人LIF基囚在COS-7细胞中能有效表达,表达产物具有较强的抑制白血病细胞生长,诱导细胞凋亡的作用。
The target human leukemia inhibitory factor (LIF) -mediated transcripts were transiently expressed in COS-7 cells and the effect of the expressed products on HL-60 leukemia cells was examined. Methods The pcDNA3.0-LIF eukaryotic expression plasmid was transfected into COS-7 cells by lipofection. The effects of COS-7 cells on HL-60 cells were studied by morphological observation, MTT and FCM. Results The human LIF gene was efficiently expressed in COS-7 cells. The expressed product could inhibit the growth and induce the apoptosis of HL-60 leukemia cells. Conclusions The human LIF-based inducible gene can express effectively in COS-7 cells. The expressed product has a strong inhibitory effect on the growth of leukemia cells and the induction of apoptosis.
