目的探讨银杏叶提取物(EGb)对糖尿病大鼠肾皮质Bcl-2和Bax表达的影响。方法 50只雄性Wistar大鼠随机分为正常对照组、糖尿病模型组、胰岛素治疗组、EGb低剂量治疗组和EGb高剂量治疗组,每组10只。糖尿病模型组、胰岛素治疗组、EGb低剂量治疗组和EGb高剂量治疗组大鼠腹腔注射链脲佐菌素(50 mg·kg-1)建立糖尿病模型,胰岛素治疗组、EGb低剂量治疗组和EGb高剂量治疗组大鼠给予不同的药物治疗3个月后,以免疫组织化学法和Western blot法检测大鼠肾皮质Bcl-2和Bax蛋白表达。结果免疫组织化学及Western blot结果表明,与正常对照组比较,糖尿病模型组大鼠肾皮质Bcl-2表达下降(P<0.01)、Bax表达上升(P<0.01);与糖尿病模型组比较,EGb低、高剂量治疗组大鼠肾皮质Bcl-2表达上升(P<0.05、P<0.01),EGb低剂量治疗组大鼠肾皮质Bax表达下降不明显(P>0.05),EGb高剂量治疗组大鼠肾皮质Bax表达显著下降(P<0.05)。结论 EGb可能通过升高糖尿病大鼠肾皮质Bcl-2表达及抑制Bax表达起到治疗作用。
Objective To investigate the effect of Ginkgo biloba extract (EGb) on the expression of Bcl-2 and Bax in renal cortex of diabetic rats. Methods Fifty male Wistar rats were randomly divided into normal control group, diabetic model group, insulin treatment group, EGb low dose treatment group and EGb high dose treatment group, with 10 rats in each group. Diabetic model group, insulin treatment group, EGb low-dose treatment group and EGb high-dose treatment group were intraperitoneally injected with streptozotocin (50 mg · kg-1) to establish diabetic model, insulin treatment group, EGb low-dose treatment group and Three months after the EGb high-dose treatment group were given different drugs, the protein expressions of Bcl-2 and Bax in rat renal cortex were detected by immunohistochemistry and Western blot. Results Immunohistochemistry and Western blot showed that compared with normal control group, the expression of Bcl-2 in renal cortex decreased (P <0.01) and the expression of Bax increased in diabetic model group (P <0.01). Compared with diabetic model group, The expression of Bcl-2 in the renal cortex of low and high dose treatment groups was increased (P <0.05, P <0.01), while the expression of Bax in renal cortex of low and high dose treatment groups was not significantly decreased (P> 0.05) The expression of Bax in rat renal cortex decreased significantly (P <0.05). Conclusion EGb may play a therapeutic role by increasing Bcl-2 expression and inhibiting Bax expression in renal cortex of diabetic rats.
