①目的 对发现于招远市的异常血红蛋白(Hb)进行分子结构的研究。②方法 取异常Hb血样,以生理盐水洗去血浆蛋白质,再以蒸馏水处理压积红细胞以溶血,溶血液以-20℃的酸性丙酮处理提取珠蛋白,用尿素将珠蛋白解链,再经柱层析分离及纯化异常链。③结果 经羧甲基纤维素及交联葡聚糖层析分离及纯化得到异常α或β链,用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS—PAGE)检查纯度,这些异常α或β链均为一条电泳带。④结论 纯化的异常α或β链均电泳纯净。
①Objective To investigate the molecular structure of abnormal hemoglobin (Hb) found in Zhaoyuan City. ② Methods Abnormal blood samples were taken from Hb, washed with saline plasma to remove plasma proteins, and then treated with distilled water to hemolysis the hematocrit. Hemolysin was extracted with acidic acetone at -20 ℃. The globin was dissolved in urea, Chromatographic separation and purification of abnormal chains. ③ The results obtained by carboxymethyl cellulose and cross-linked dextran chromatography separation and purification of abnormal α or β chain, with sodium dodecyl sulfate - polyacrylamide gel electrophoresis (SDS-PAGE) to check the purity of these abnormalities Alpha or beta chain are an electrophoresis band. ④ Conclusion Purified abnormal α or β chain electrophoresis were pure.
