目的分离鉴定枯草芽孢杆菌PJS发酵液中的环脂肽类抗生素PJS-0。方法采用溶剂萃取、薄层色谱(TLC)、高效液相色谱法(HPLC)等分离方法对菌株PJS发酵产物的活性组分PJS-0进行分离;并通过紫外(UV)、质谱(MS)和核磁共振波谱(NMR)联合分析对其结构进行鉴定。结果从塔克拉玛干沙漠来源植物内生枯草芽孢杆菌PJS的发酵液中分离得到一个环脂肽类抗生素:PJS-0,经结构鉴定为Mojavensin A。结论本文首次从陆生芽孢杆菌中分离得到Mojavensin A,并首次用ESI+-MS/MS对其氨基酸序列进行鉴定。
Objective To isolate and identify the lipopeptide antibiotic PJS-0 in Bacillus subtilis PJS fermentation broth. Methods PJS-0, the active component of the fermentation product of PJS strain, was isolated by solvent extraction, TLC and HPLC. The components of PJS-0 were separated by ultraviolet (UV), mass spectrometry (MS) and Nuclear magnetic resonance (NMR) analysis of the structure of the joint identification. Results A lipopeptide antibiotic: PJS-0 was isolated from the fermentation broth of endophytic Bacillus subtilis PJS from the Taklimakan Desert, and identified as Mojavensin A by its structure. CONCLUSION Mojavensin A was isolated from Bacillus thuringiensis for the first time and its amino acid sequence was first identified by ESI + -MS / MS.
