目的:建立RP-HPLC法测定人血浆中美洛昔康浓度。方法:采用Symmetry C_(18)分析柱(150mm×3.9 mm,5μm),以甲醇-20mmo·L~(-1)磷酸二氢钠溶液(52:48;磷酸调pH至5.20)为流动相,流速1.2mL·min~(-1)。血浆样品以卡马西平作内标,酸化后用氯仿提取并浓缩进样,检测波长为271nm。结果:标准曲线线性范围为0.02~2.0 mg·L~(-1),血浆美洛昔康萃取回收率为87.6%~92.9%,加样回收率为97.0%~99.4%,日内RSD为1.3%~1.8%,日间RSD为3.4%~5.8%。结论:本法简便,准确,重现性好,用于美洛昔康人体药动学研究血药浓度测定取得良好结果。
Objective: To establish a RP-HPLC method for the determination of meloxicam in human plasma. METHODS: Symmetry C_ (18) analytical column (150 mm × 3.9 mm, 5 μm) was used with mobile phase of methanol-20 mmo · L -1 sodium dihydrogen phosphate solution (52:48; Flow rate 1.2mL · min ~ (-1). Plasma samples were taken as internal standard with carbamazepine, acidified and extracted with chloroform and concentrated. The detection wavelength was 271 nm. Results: The linear range of standard curve was 0.02-2.0 mg · L -1, the recovery of meloxicam in plasma was 87.6% -92.9%, the recovery of sample was 97.0% -99.4%, and the intraday RSD was 1.3% ~ 1.8%, daytime RSD was 3.4% ~ 5.8%. Conclusion: The method is simple, accurate and reproducible. The results of the determination of blood concentration of meloxicam in human pharmacokinetics have achieved good results.
