目的 研究紫外线 (UVB)诱导成纤维细胞 (NIH3T3 )凋亡时胞浆pH值的变化。方法 利用粘附式细胞仪(ACAS -5 70 )测定UVB诱导NIH3T3细胞凋亡时 ,胞浆pH值的变化。结果 NIH3T3细胞经UVB照射后 ,胞内pH值会逐渐升高 ,并在 1 6h左右达到最大值 ,2 5min后恢复正常 ,在细胞内外钙被螯合后 ,不管UVB照射与否 ,胞内pH值无变化。结论 UVB照射导致细胞凋亡时 ,胞液呈碱性 ,pH值的变化参与了细胞凋亡的调控 ;细胞内外Ca2 + 水平影响胞浆pH值的变化 ,胞浆Ca2 + 与胞浆pH值的变化存在一定的相关关系。
Objective To study the change of cytoplasm pH value induced by ultraviolet (UVB) -induced fibroblasts (NIH3T3) apoptosis. Methods The change of cytoplasm pH value in UVB-induced apoptosis of NIH3T3 cells was detected by adherent cytometer (ACAS -570). Results After UVB irradiation, the intracellular pH value of NIH3T3 cells increased gradually and reached a maximum at about 16 h and returned to normal after 25 min. After intracellular calcium chelation, no matter the UVB irradiation or not, the intracellular pH No change in value. CONCLUSION: UVB irradiation results in alkaline cytosolic fluid and changes in pH value are involved in the regulation of apoptosis. Intracellular Ca 2+ levels affect the changes of pH in cytoplasm. The changes of cytosolic calcium and cytoplasmic pH There is a certain correlation between changes.
