目的 探讨细胞周期抑制因子p16在细胞衰老中的作用。 方法 利用逆转录病毒载体将p16基因转染入人胚肺二倍体成纤维细胞 2BS中 ,获得高表达 ,检测其对 2BS细胞衰老的影响。 结果 与对照组细胞相比 ,p16基因转染后 ,2BS细胞生长速率下降了约 5 0 % ,细胞周期阻滞于G1期 ,细胞对生长因子剌激的反应性下降了 79 4% ,细胞形态呈衰老细胞样变化。 结论 p16在人二倍体成纤维细胞的衰老过程中起促进作用。
Objective To investigate the role of cell cycle inhibitor p16 in cell senescence. Methods The p16 gene was transfected into human fetal lung diploid fibroblasts 2BS by retroviral vector and the expression of p16 gene was observed. The effect of p16 gene on the senescence of 2BS cells was observed. Results Compared with the control group, the growth rate of 2BS cells decreased by about 50% after transfection of p16 gene and the cell cycle arrest in G1 phase. The reactivity of cells to growth factor stimulation decreased by 79.4% Senescent cell-like changes. Conclusion p16 plays an important role in the senescence of human diploid fibroblasts.
