目的检测缺氧缺血性脑损伤(hypoxic-ischemic brain damage,HIBD)后Foxg1基因及脑皮质特定基因(Foxp2,ER81,Otx1,Foxp1,RORβ,Reelin)的动态变化。方法采用Rice法制作新生鼠HIBD动物模型,假手术组作为对照组,分别在术后第3、7、14、28、56d取右侧脑半球。半定量RT-PCR检测各基因表达水平。结果HIBD后Foxg1基因表达水平模型组3d较对照组低(P<0.05),14、56d表达水平均较对照组高(P<0.05);Reelin基因表达水平在术后7d及28d均较对照组高(P<0.05);Foxp2基因在7、14、56d表达水平均较对照组高(P均<0.05)。Otx-1基因在术后7、14、28、56d表达水平均较对照组高(P均<0.05),模型组与对照组Otx-1的表达均呈下降趋势;RORβ、Foxp1、ER81表达趋势尚不能肯定。结论HIBD后Foxg1、Reelin及Foxp2表达水平呈上升趋势,Otx1呈下调趋势,RORβ、Foxp1、ER81表达趋势尚不能肯定,Foxg1基因表达变化与脑损伤后大脑结构自身修复的具体机制有待进一步研究。
Objective To investigate the dynamic changes of Foxg1 and cortex specific genes (Foxp2, ER81, Otx1, Foxp1, RORβ, Reelin) after hypoxic-ischemic brain damage (HIBD). Methods Newborn HIBD animal model was made by the method of Rice and the sham operation group was taken as the control group. The right hemisphere was taken on the 3rd, 7th, 14th, 28th and 56th day respectively. Semi-quantitative RT-PCR detection of gene expression levels. Results The expression of Foxg1 in HIBD group was lower than that in control group (P <0.05), and the expression level of Foxg1 in 14th and 56th days was higher than that in control group (P <0.05). The expression level of Reelin gene in 7d and 28d after operation were higher than those in control group (P <0.05). The expression level of Foxp2 in the 7th, 14th and 56th day was higher than that in the control group (all P <0.05). The expression of Otx-1 mRNA at 7, 14, 28 and 56 days after operation was significantly higher than that of the control group (all P <0.05), and the expression of Otx-1 in the model group and the control group showed a decreasing trend. The expression of RORβ, Foxp1 and ER81 Still not sure. CONCLUSION: The expression of Foxg1, Reelin and Foxp2 in HIBD are up-regulated and the expression of Otx1 is down-regulated. The expression of RORβ, Foxp1 and ER81 remains uncertain. The specific mechanism of Foxg1 gene expression and brain tissue repair after brain injury remains to be further studied.
