目的探讨24例宣威肺腺癌病人癌组织中miR-501-5p的表达与临床病理因素之间的关系及miR-501-5p在宣威地区肺腺癌中的作用。方法收集24例宣威地区肺腺癌患者的癌组织及相应的正常肺标本,提取组织中的微小RNA(microRNA),采用QPCR技术,microRNAs芯片扫描,检测miR-501-5p在肺癌组织和相应正常肺组织的表达量。使用χ2检验对miR-501-5P在宣威地区肺腺癌标本的表达量与临床特征因素(年龄,性别,肺癌分期及术前CEA值)进行统计学分析,运用多重回归分析miR-501-5p表达与临床特征(年龄,肺癌分期及术前CEA值)的关系。结果通过对24对宣威地区肺腺癌配对样本进行microRNA芯片检测和QPCR验证均提示:miR-501-5p在宣威地区肺腺癌中呈上调(P<0.01)。mciroRNA芯片结果(癌vs肺组织)表达差异倍数:3倍(P<0.05,FDR:0.07)。qPCR验证结果(癌vs正常肺组织)表达倍数(x±s):5.702±4.626。χ~2检验芯片miR-501-5p表达量与临床特征结果显示与年龄(f=7.168,P=0.014),肺癌TNM分期(f=36.627,P<0.01)及术前CEA值f=30.045,P<0.01)关系密切;但与性别(f=3.612,P=0.071)无关。在多重回归分析结果提示:miR-501-5p表达量与患者年龄,肺癌TNM分期、血清CEA(癌胚抗原)均呈正相关(P<0.05)。结论 miR-501-5p在宣威地区肺腺癌标本中呈高表达,其与年龄,临床肺癌TNM分期、血清CEA呈显著相关,可能参与宣威地区肺腺癌的发生发展。
Objective To investigate the relationship between the expression of miR-501-5p and the clinicopathological factors in 24 cases of Xuanwei lung adenocarcinoma and the role of miR-501-5p in lung adenocarcinoma in Xuanwei region. Methods Twenty-four patients with lung adenocarcinoma in Xuanwei district were recruited and their corresponding normal lung specimens were collected. MicroRNAs (microRNAs) were extracted from the tissues of patients with lung adenocarcinoma in Xuanwei area. QPCR and microRNAs chip scanning were performed to detect the expression of miR-501-5p in lung cancer tissues and their corresponding Normal lung tissue expression levels. The expression of miR-501-5P in lung adenocarcinoma specimens of Xuanwei area and clinical characteristics (age, sex, stage of lung cancer and preoperative CEA value) were analyzed by Chi-square test. Multiple regression analysis was used to analyze the expression of miR-501- 5p expression and clinical features (age, stage of lung cancer and preoperative CEA value). Results MicroRNA microarray detection and QPCR analysis of 24 pairs of lung adenocarcinoma matched samples in Xuanwei region indicated that miR-501-5p was upregulated in lung adenocarcinoma in Xuanwei region (P <0.01). mciroRNA chip results (cancer vs. lung tissue) Multiple fold difference: 3-fold (P <0.05, FDR: 0.07). qPCR test results (cancer vs normal lung tissue) expression fold (x ± s): 5.702 ± 4.626. The expression and clinical features of miR-501-5p in χ ~ 2 test showed that the expression level of miR-501-5p and clinical features in patients with lung cancer were significantly higher than those in patients with age (f = 7.168, P = 0.014), TNM stage (f = 36.627, P <0.01), but not with gender (f = 3.612, P = 0.071). The results of multiple regression analysis indicated that the expression of miR-501-5p was positively correlated with patient’s age, TNM stage of lung cancer, serum CEA (carcinoembryonic antigen) (P <0.05). Conclusions miR-501-5p is highly expressed in lung adenocarcinoma specimens in Xuanwei area, which is significantly correlated with age, TNM stage of clinical lung cancer and serum CEA, which may be involved in the occurrence and development of lung adenocarcinoma in Xuanwei area.
