目的探讨C型凝集素16A(CLEC16A)在中枢神经系统(CNS)炎症中的表达变化及细胞定位。方法应用SD大鼠侧脑室内注射脂多糖(LPS)建立脑炎症模型,利用PCR及Western blot检测术后大脑皮层中CLEC16A在mRNA水平及蛋白水平的表达变化;免疫荧光检测CLEC16A在侧脑室内注射LPS后的定位、细胞分布及细胞的状态。结果在大鼠脑炎症模型中CLEC16A mRNA和蛋白水平均有明显变化,分别在注射后9或12 h开始增加,并一直维持在较高水平。免疫荧光双标发现表达较高CLEC16A主要定位与大鼠脑中的星形胶质细胞及神经元中。同时检测脑中星形胶质细胞活化的情况发现表达增多的CLEC16A主要定位于增殖的星形胶质细胞。结论CLEC16A可能参与星形胶质细胞的增殖,在神经系统炎症反应过程中发挥着重要的作用。
Objective To investigate the expression and location of C-type lectin-16A (CLEC16A) in the inflammation of central nervous system (CNS). Methods Cerebral inflammation model was established by intracerebroventricular injection of lipopolysaccharide (LPS) in SD rats. The expression of CLEC16A mRNA and protein in cerebral cortex were detected by PCR and Western blot. The expression of CLEC16A was detected by immunofluorescence in lateral ventricle Location after LPS, cell distribution and cell status. Results The mRNA and protein levels of CLEC16A in rat model of cerebral encephalopathy were significantly changed, which began to increase at 9 or 12 h after injection and maintained at a high level. Immunofluorescence double labeling was found to be expressed in higher CLEC16A mainly localized in astrocytes and neurons in the rat brain. Simultaneous detection of astrocyte activation in the brain revealed that the increased expression of CLEC16A predominantly localized in proliferating astrocytes. Conclusion CLEC16A may be involved in the proliferation of astrocytes and play an important role in the nervous system inflammatory response.
