目的:探讨脑膜瘤组织中基质金属蛋白酶-2、9(MMP-2、MMP-9)及其特异性金属蛋白酶组织抑制剂-1、2(TIMP-1、TIMP-2)基因mRNA的表达及其与肿瘤侵袭和转移性的关系。方法:采用逆转录聚合酶链反应(RT-PCR)法半定量检测36例不同病理类型脑膜瘤组织中MMP-2、MMP-9及TIMP-1、TIMP-2基因mRNA的表达。结果:MMP-2及TIMP-2mRNA在36例脑膜瘤组织中均有表达,除MMP-2在良性组与非典型组间比较差异无统计学意义外(P>0.05),其余各组间比较差异均有统计学意义,P均<0.05。MMP-9及TIMP-1mRNA在36例脑膜瘤组织中也均有表达,各组间比较差异均有统计学意义,P均<0.05。MMP-2/TIMP-2比值在良性组与非典型组间比较差异无统计学意义,P>0.05,而恶性组与其他两组间比较差异均有统计学意义,P均<0.05。MMP-9/TIMP-1比值在3组间比较差异均有统计学意义,P均<0.05。MMPs/TIMPs比值随着肿瘤恶性程度的增加而有逐渐增大的趋势。结论:MMPs/TIMPs基因表达的比值平衡失调在脑膜瘤的侵袭和转移性方面具有重要作用。
Objective: To investigate the mRNA expression of matrix metalloproteinase-2, 9 (MMP-2, MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) Its relationship with tumor invasion and metastasis. Methods: The mRNA expressions of MMP-2, MMP-9, TIMP-1 and TIMP-2 in 36 cases with different pathological types of meningiomas were detected by RT-PCR. Results: MMP-2 and TIMP-2 mRNA were all expressed in 36 cases of meningiomas, except MMP-2 in benign group and non-typical group, there was no significant difference (P> 0.05). The other groups were compared The differences were statistically significant, P <0.05. MMP-9 and TIMP-1 mRNA were also expressed in 36 cases of meningiomas, the difference was statistically significant between groups, P <0.05. There was no significant difference in MMP-2 / TIMP-2 ratio between benign group and non-typical group (P> 0.05), while there was significant difference between malignant group and other two groups (P <0.05). MMP-9 / TIMP-1 ratio in the three groups were statistically significant differences, P <0.05. The ratio of MMPs / TIMPs tended to increase with the increase of malignancy. Conclusion: The imbalanced ratio of MMPs / TIMPs gene expression plays an important role in the invasion and metastasis of meningioma.
