目的:研究金雀异黄素对人脐静脉内皮细胞(hU-VEC)生长的影响及机制。方法:培养并鉴定hUVEC;用不同浓度金雀异黄素(0·1、1、10和50μmol)对hUVEC作用24h;先用MTT比色法观察细胞存活率、流式细胞术检测细胞DNA含量及细胞周期观察其对hUVEC生长的影响,流式细胞术检测凋亡相关蛋白Fas、Bcl-2的表达,探讨金雀异黄素诱导hU-VEC增殖的可能机制。结果:金雀异黄素能诱导hUVEC的增殖(P<0·05或P<0·01),其效应随浓度的增加而增强;细胞周期显示S期细胞明显增加;金雀异黄素促进Bcl-2的表达,抑制Fas的表达。结论:金雀异黄素能诱导hUVEC增殖,其机制可能与上调Bcl-2及下调Fas的表达有关。
Objective: To study the effect of genistein on the growth of human umbilical vein endothelial cells (hU-VEC) and its mechanism. METHODS: hUVECs were cultured and identified; hUVECs were treated with different concentrations of genistein (0·1, 1, 10, and 50 μmol) for 24 h; cell survival was measured by MTT colorimetry, and DNA content was measured by flow cytometry. The cell cycle was used to observe its effect on hUVEC growth. Flow cytometry was used to detect the expression of apoptosis-related proteins Fas and Bcl-2, and to explore the possible mechanism of genistein-induced hU-VEC proliferation. RESULTS: Genistein could induce proliferation of hUVEC (P<0.05 or P<0.01), and its effect increased with increasing concentration; cell cycle showed a significant increase in cells in S phase; genistein promoted Bcl- Expression of 2 inhibits the expression of Fas. Conclusion: Genistein can induce hUVEC proliferation, which may be related to up-regulation of Bcl-2 and down-regulation of Fas expression.
