目的:研究四川江油、陕西汉中及云南丽江等附子主产区黑顺片的高效液相色谱指纹图谱,为建立附子的行业质量标准提供依据。方法:利用HPLC方法,色谱条件Alltimate C18色谱柱(4.6 mm×250 mm,5μm),柱温35℃,流动相40mmol.L-1醋酸铵(氨水调pH 10.00)-乙腈梯度洗脱,流速1.0 mL.min-1,检测波长为240 nm。对不同主产地的22批黑顺片进行研究,采用药典委员会颁布的中药色谱指纹图谱相似度评价系统2004A和聚类分析对指纹图谱进行了分析。结果:22批黑顺片有11个共有峰,江油地区(道地产区)的黑顺片之间的相似度均大于0.900,非道地产区与道地产区的相似度小于0.900。结论:该方法精密度、重复性和稳定性良好,可结合黑顺片含量测定用于全面控制附子炮制品(黑顺片)的质量。
OBJECTIVE: To study the HPLC fingerprint of Heshun tablet in the main producing areas of Fuzi, Sichuan Jiangyou, Hanzhong of Shaanxi Province and Lijiang of Yunnan Province, and to provide basis for the establishment of quality standards of aconite. Methods: HPLC method was used to elute all the analytes on a C18 column (4.6 mm × 250 mm, 5 μm) with a column temperature of 35 ° C and a mobile phase of 40 mmol.L-1 ammonium acetate (ammonia water, pH 10.00) mL.min-1, the detection wavelength was 240 nm. Twenty-two batches of Heshun tablets from different main producing areas were studied. The fingerprints were analyzed by using the similarity evaluation system of chromatographic fingerprints of traditional Chinese medicine (TCM) 2004A and the cluster analysis promulgated by the Pharmacopoeia Committee. Results: There were 11 common peaks in 22 batches of Heshun tablet and the similarities between Heshun tablets in Jiangyou region (Tao real estate area) were both greater than 0.900. The similarities between non-real estate areas and real estate areas were less than 0.900. Conclusion: The method has good precision, repeatability and stability. It can be used to control the quality of the aconite product (Heshun tablet) in combination with the determination of the Heshun Tablet.
