目的研究黄芪多糖抗衰老的作用机制。方法β-半乳糖苷酶(SA-β-gal)及吖啶橙(AO)的荧光染色分析黄芪多糖对斑马鱼胚胎细胞衰老及凋亡的影响;RT-PCR检测与衰老相关基因的表达。结果当黄芪多糖质量浓度为0.25 mg.mL-1时,可以延缓斑马鱼细胞凋亡;增强斑马鱼端粒酶基因(TERT)基因的表达,降低bax、p21、p53基因的表达。结论黄芪多糖具有改善细胞复制性衰老的作用,其分子机制可能与上调TERT和下调p21、p53和bax基因的表达,从而起到一定的抗衰老作用。
Objective To study the anti-aging mechanism of Astragalus polysaccharides. Methods The effects of Astragalus polysaccharides (APS) on the senescence and apoptosis of zebrafish embryos were analyzed by fluorescent staining of β-galactosidase (SA-β-gal) and acridine orange (AO). The expression of aging related genes was detected by RT- Results When astragalus polysaccharide was 0.25 mg.mL-1, the apoptosis of zebrafish cells could be delayed, the expression of telomerase gene (TERT) gene and the expression of bax, p21 and p53 genes were enhanced. Conclusion Astragalus polysaccharides can improve the reproductive cell senescence, and its molecular mechanism may be related to the up-regulation of TERT and the down-regulation of the expression of p21, p53 and bax gene, which play a certain anti-aging effect.
