目的 :观察非选择性一氧化氮合酶抑制剂 NG 硝基左旋精氨酸 (L NNA)对缺血性海马神经元凋亡的影响 ,探讨一氧化氮 (NO)在缺血性神经元凋亡中的作用。方法 :2 7只雄性 Wistar大鼠分为假手术组、生理盐水治疗组和 L NNA治疗组。采用 4血管关闭法制成全脑缺血模型 ,利用 DNA10 %聚丙烯酰胺凝胶电泳法和原位末端标记技术 ,观察计数全脑缺血 10分钟再灌注 72小时海马区神经元凋亡情况。结果 :全脑缺血 10分钟再灌注 72小时海马区神经元出现凋亡〔凋亡细胞 (39.0 0± 2 .74)个〕,L NNA可抑制缺血性神经元凋亡〔凋亡细胞 (31.80± 3.71)个 ,P<0 .0 5〕。结论 :NO能诱导缺血性神经元凋亡 ,小剂量反复使用 L NNA能有效抑制缺血性海马区神经元凋亡
Objective: To observe the effect of non-selective nitric oxide synthase inhibitor L-NNA on the apoptosis of ischemic hippocampal neurons and to explore the role of nitric oxide (NO) in ischemic neuronal apoptosis The role of death. Methods: Twenty seven male Wistar rats were divided into sham operation group, saline control group and L NNA treatment group. The model of global cerebral ischemia was established by 4-vessel occlusion method. The apoptosis of hippocampal neurons was observed by DNA 10% polyacrylamide gel electrophoresis and in situ end-labeling technique for 72 hours after reperfusion for 10 minutes. Results: Apoptosis of neurons in hippocampus (apoptotic cells (39.0 0 ± 2.74)] was observed after reperfusion for 10 min at 10 min after global cerebral ischemia. L NNA could inhibit the apoptosis of ischemic neurons [apoptotic cells ( 31.80 ± 3.71), P <0.05. CONCLUSION: NO can induce the apoptosis of ischemic neurons. L NNA can effectively inhibit the apoptosis of ischemic hippocampal neurons in low dose
