不同毒性等级的化学品对线虫感知行为的影响

目的本研究测定并评价化学品作用于模式生物线虫后对其感知行为的毒性影响,为今后将模式生物线虫应用于化学品毒性研究和评价奠定基础。方法以GHS III到VI毒性等级的化学品为研究对象,每等级中选取3~8个化学品,对秀丽隐杆线虫(Nematode Caenorhabditis elegans,以下简称线虫)进行6 h亚急性染毒,采用趋避指数和趋向指数,测定并评价化学品作用于模式生物线虫后对其感知行为的毒性影响,分析亚急性染毒条件下上述指数测定值的变化与化学品GHS毒性分级间的联系,以及与本课题组前期研究获得的线虫急性毒性指标(24 h LC50)之间的内在关联性,探讨以上述指数作为化学品非致死性评价初始检测指标的可行性,并据此提出今后化学品的线虫亚急性毒性检测的推荐染毒剂量范围。结果化学品作用于线虫的趋避指数和趋向指数与其GHS分级、24 h LC50有一定关联:GHS级别越高,出现趋避阳性的概率越低,且趋避阳性的最高剂量值也越大;GHS级别越高,出现趋向阳性的概率越高,且趋向阳性的最高剂量值越大。结论线虫趋避指数和趋向指数与化学品毒性大小有一定的关联性,推荐将这两个指数作为研究化学品的线虫亚急性毒性检测的初始检测指标。基于本实验结果推荐今后线虫的亚急性毒性实验的最高染毒剂量为其24 h LC50的1/12~1/2。 OBJECTIVE: This study determined and evaluated the toxic effects of chemicals acting on the biological behavior of the model organism C. elegans, and laid the foundation for future application of the model organism in research and evaluation of chemical toxicity. Methods The chemicals of GHS III to VI toxicity level were selected as the research objects. Three to eight chemicals were selected for each grade. Nematode Caenorhabditis elegans (subgenus nematode) was sub-acutely exposed for 6 hours. Index and trend index, determine and evaluate the toxic effects of chemicals on the perceived behavior of the model organism, and analyze the relationship between the change of the above indices and chemical GHS toxicity classification under subacute exposure conditions, The intrinsic relevance of the acute toxicity index of nematodes (24 h LC50) obtained from the previous study of the research group and the feasibility of using the above index as the initial detection index of non-lethal chemical evaluation, and to propose the future line of nematode Acute toxicity testing recommended dose range. Results The evasion index and trend index of chemicals acting on nematodes correlated with their 24 h LC50. The higher the GHS level, the lower the probability of avoiding positive and the higher the maximum dose to avoid positive. The higher the GHS level, the higher the GHS level High, the higher the probability of positive, and the higher the highest dose positive trend. Conclusions The nematode repellent index and trend index are related to the toxicity of chemicals. It is recommended to use these two indexes as the initial detection index for the detection of subacute toxicity of nematodes in chemical products. Based on the results of this experiment, the highest exposure dose of sub-acute toxicities to nematodes in the future was 1/12 to 1/2 of its 24 h LC50.
