目的探讨右归饮治疗激素性股骨头坏死的可能作用机理。方法 30只SD大鼠随机分成正常对照组、右归饮组、激素模型组,每组10只。激素模型组和右归饮组采用腹腔内注射大肠杆菌内毒素联合肌肉注射甲泼尼松龙琥珀酸钠建立激素性股骨头坏死模型,正常对照组给予等量生理盐水注射。右归饮组在造模的同时每天给予右归饮20 g/(kg·d)灌胃,连续8周,激素模型组和正常对照组予等容积生理盐水灌胃。观察各组大鼠一般情况,于第4、8周末行骨X线及骨密度检测,第8周末测定血清钙、磷水平。结果与正常对照组比较,激素模型组和右归饮组大鼠第2周开始至第8周体重均明显降低(P<0.05);从第3周至第8周右归饮组大鼠体重较激素模型组明显升高(P<0.05)。激素模型组大鼠第4、8周股骨干、股骨头、脊柱骨密度水平均较正常对照组明显下降,而右归饮组第4周时股骨干、股骨头和第8周时股骨干、股骨头、脊柱骨密度水平较激素模型组明显升高(P<0.01)。与正常对照组比较,右归饮组和激素模型组血清钙、磷水平均降低(P<0.01);与激素模型组比较,右归饮组血清钙、磷水平升高(P<0.01)。结论右归饮能明显提高激素性股骨头坏死模型大鼠的骨密度,其机理可能与改善血清钙、磷水平有关。
Objective To explore the possible mechanism of You Gui Yin in treating steroid-induced femoral head necrosis. Methods Thirty SD rats were randomly divided into normal control group, right-handed drink group and hormone model group, with 10 rats in each group. Hormone model group and Youguiyin group were given intraperitoneal injection of E. coli endotoxin intramuscular injection of methylprednisolone sodium succinate to establish a model of steroid-induced osteonecrosis, normal control group given an equal volume of saline injection. Youguiyin group was treated with Yougui Decoction 20 g / (kg · d) daily for 8 weeks, while the model group and normal control group were given intragastric administration of normal saline. The general conditions of rats in each group were observed. X-ray and bone mineral density were measured at the end of the 4th and 8th week. Serum calcium and phosphorus levels were measured at the end of the 8th week. Results Compared with the normal control group, the body weight of the rats in the model group and the right-handed drink group decreased significantly from the second week to the eighth week (P <0.05); from the third week to the eighth week, The hormone model group was significantly higher (P <0.05). At the 4th and 8th week in the hormone model group, the BMD of the femoral head, femoral head and spine decreased significantly in the 4th and 8th weeks in the model group, compared with the femoral shaft, The bone mineral density of the femoral head and spine increased significantly compared with the model group (P <0.01). Compared with the normal control group, the levels of serum calcium and phosphorus in Yougui Yin group and the model group were lower than those in the normal control group (P <0.01). Compared with the model group, the level of serum calcium and phosphorus in Yougui Yin group was increased (P <0.01). Conclusion Yougui Yin can significantly increase the bone mineral density in steroid-induced osteonecrosis of the femoral head in rats, and its mechanism may be related to the improvement of serum calcium and phosphorus levels.
