本研究旨在探讨Cu2+浓度梯度、胁迫时间对禾花鲤(Procypris mterus)血液和肝脏的过氧化氢酶(catalase,CAT)活性的影响,为禾花鲤的重金属胁迫研究及水污染生物监测提供理论参考依据,进一步指导禾花鲤的健康养殖。采用可见光法测定了不同Cu2+浓度(0、0.067、0.084、0.101、0.118和0.134 mg.L-1)和胁迫时间(0、1、5和11 d)下禾花鲤CAT活性的变化。结果显示,在试验的各个持续时间段(1、5和11 d),低Cu2+浓度处理组(0.067 mg.L-1)血液及肝脏的CAT活性高于其它各处理组,且活性随着胁迫时间的延长而增加。而中、高Cu2+浓度处理组(0.084、0.101、0.118和0.134 mg.L-1)血液和肝脏中的CAT活性则随着Cu2+浓度的升高和胁迫时间的延长被显著抑制(P<0.05),活性变化在5 d后趋于稳定。试验表明鱼体过氧化氢酶的活性受水体中Cu2+浓度的影响,低Cu2+浓度能诱导鱼体血液和肝脏中的CAT活性增加,但随胁迫的加剧,CAT活性受到抑制。
The purpose of this study was to investigate the effects of Cu2 + concentration gradient and stress time on the catalase (CAT) activity in the blood and liver of Procypris mterus, and to provide a basis for the study of heavy metal stress and bio-monitoring of water pollution in grass carp Theoretical reference, to further guide the healthy breeding of grass carp. The changes of CAT activity in grass carp (Cyprinus carpio L.) under different concentrations of Cu2 + (0,0.067,0.084,0.101,0.118 and 0.134 mg.L-1) and stress time (0, 1, 5 and 11 days) were measured by visible light method. The results showed that the CAT activity in the blood and liver of low Cu2 + -treated group (0.067 mg.L-1) was higher than that of other treatment groups at various durations (1, 5 and 11 d) The increase of time. However, the activities of CAT in blood and liver of medium and high Cu2 + -treated groups (0.084,0.101,0.118 and 0.134 mg.L-1) were significantly inhibited with the increase of Cu2 + concentration and prolongation of stress time (P <0.05) , The change of activity tended to be stable after 5 days. The results showed that the activity of catalase in fish was affected by the concentration of Cu2 + in the water. The low Cu2 + concentration could induce the increase of CAT activity in the blood and liver, but the CAT activity was inhibited as the stress increased.
