目的建立HPLC-ELSD法分析油菜蜂花粉中6种脂肪酸,并测定其含量。方法色谱柱Agilent ZORBAX SB-C18(4.6 mm×250 mm,5μm);流动相:甲醇-1%乙酸水梯度洗脱,流速1.2 mL·min-1,柱温25℃;检测器参数:增益:6.0,漂移管温度:30℃,载气:氮气,压力:0.3 MPa。结果十四烷酸、亚麻酸、亚油酸、软脂酸、油酸、十八烷酸分离度均在1.5以上,加样回收率均位于95.3%~103.7%,相对标准偏差(n=6)分别为2.2%,2.0%,1.0%,1.4%,1.4%,2.3%。结论该方法简便、准确、重复性好,适合油菜蜂花粉原药材中高含量脂肪酸成分的分析测定。
OBJECTIVE To establish a HPLC-ELSD method for the determination of six fatty acids in rape bee pollen and determine its content. Methods Column Agilent ZORBAX SB-C18 (4.6 mm × 250 mm, 5 μm) was used. The mobile phase was eluted with a gradient of methanol and 1% acetic acid. The flow rate was 1.2 mL · min-1 and the column temperature was 25 ℃. 6.0, drift tube temperature: 30 ° C, carrier gas: nitrogen, pressure: 0.3 MPa. Results The myristic acid, linolenic acid, linoleic acid, palmitic acid, oleic acid and octadecanoic acid were all above 1.5, and the recoveries were in the range of 95.3% ~ 103.7%. The relative standard deviations (n = 6 ) Were 2.2%, 2.0%, 1.0%, 1.4%, 1.4%, 2.3% respectively. Conclusion The method is simple, accurate and reproducible. It is suitable for the determination of high content of fatty acid in rape bee pollen raw material.
