目的:通过观察明月草水提液对DPP-4、α-葡萄糖苷酶的抑制作用,为该药防治糖尿病提供实验依据。方法:以DPP-4酶、缓冲液、底物建立DPP-4抑制剂的体外筛选体系,对明月草水提液进行抑制实验,采用发色底物法测定吸光度(OD),计算DPP-4抑制率及IC50值。以蔗糖为底物建立α-葡萄糖苷酶活性抑制模型,采用葡萄糖氧化酶法测定明月草水提液对α-葡萄糖苷酶的抑制作用,计算其抑制率和IC50值。结果:明月草具有轻度DPP-4抑制作用,其IC50值为1199.97mg/L。明月草具有α-葡萄糖苷酶抑制作用,其IC50值为64.12mg/L。结论:明月草水提液可一定程度地抑制DPP-4及α-葡萄糖苷酶活性。
OBJECTIVE: To provide an experimental basis for the prevention and treatment of diabetes mellitus by observing the inhibitory effect of the aqueous extract of Melissa officinalis on DPP-4 and α-glucosidase. Methods: The in vitro screening system of DPP-4 inhibitor was established by DPP-4 enzyme, buffer and substrate. The inhibitory effect of DPP-4 inhibitor on the aqueous extract of DPP-4 was determined. The absorbance (OD) Inhibition rate and IC50 value. Inhibitory activity of α-glucosidase was determined by using sucrose as substrate. The inhibitory rate and IC50 value of α-glucosidase were determined by glucose oxidase method. Results: Mild moon grass had a mild DPP-4 inhibition, the IC50 value of 1199.97mg / L. Mingyue grass has α-glucosidase inhibition, the IC50 value of 64.12mg / L. CONCLUSION: The extract of Melasmaria gracilis could inhibit the activity of DPP-4 and α-glucosidase to some extent.
