目的:建立糙叶败酱药材中异白花败酱醇的定性和定量分析方法。方法:采用薄层色谱(TLC)法定性鉴别糙叶败酱药材中的异白花败酱醇;采用反相高效液相色谱法测定其含量,色谱柱为Agilent Eclipse plus C18(250mm×4.6mm,5μm),流动相为乙腈-磷酸水溶液(pH2.5)=7:93,流速为1.0mL.min-1,检测波长为210nm,柱温为30℃。结果:糙叶败酱药材中异白花败酱醇的TLC鉴别特征明显,专属性好;异白花败酱醇的进样浓度在0.2~1.2mg.mL-1范围内与峰面积积分值呈良好线性关系(r=0.9997),平均加样回收率为98.8%,RSD=2.84%(n=9)。结论:所建分析方法准确、可靠,可用于糙叶败酱的质量控制。
Objective: To establish a qualitative and quantitative analysis method of Patrinia. Methods: TLC method was used for the qualitative identification of palmatine in Paeonia lactiflora. The content was determined by RP-HPLC with Agilent Eclipse plus C18 (250 mm × 4.6 mm, 5μm). The mobile phase was acetonitrile-phosphoric acid (pH2.5) = 7:93, the flow rate was 1.0mL.min-1, the detection wavelength was 210nm and the column temperature was 30 ℃. Results: The TLC identification characteristics of Patrinia purpurea were obvious and the specificity was good. The injection concentration of Patrinia sativa was good with the peak area in the range of 0.2-1.2 mg.mL-1 The linear relationship (r = 0.9997), the average recovery was 98.8%, RSD = 2.84% (n = 9). Conclusion: The established analytical method is accurate and reliable, and can be used for quality control of Cassia.
