目的:研究糖皮质激素性骨质疏松症大鼠骨组织中BMP6/BMP7的mRNA和蛋白表达,揭示糖皮质激素骨质疏松症的发病机理,阐述纳米钙补肾中药的调节作用机制。方法:用糖皮质激素注射的方法复制骨质疏松症模型,用纳米钙及具有益肾填精、补钙壮骨作用的补肾中药高、低剂量对实验大鼠治疗8周,以骨疏康颗粒、盖天力钙片作为阳性对照组,正常大鼠和模型空白组为空白对照组。用PCR法、Western印迹法检测骨质疏松症大鼠骨组织中BMP6/BMP7mRNA和蛋白表达。结果:正常大鼠骨组织可以在基因、蛋白水平表达BMP6及BMP7,模空组大鼠骨组织中的BMP6/BMP7 mRNA和蛋白表达水平明显的下降。纳米钙补肾中药高、低剂量组、盖天力组、骨疏康组,用药8周后,与模空组相比较,均可上调骨组织中BMP6/BMP7 mRNA和蛋白的表达水平。结论:纳米钙补肾中药通过调控大鼠骨组织中BMP6/BMP7 mRNA和蛋白表达,抑制骨吸收,促进骨形成,从而对糖皮质激素性骨质疏松症有一定的防治作用。
OBJECTIVE: To study the mRNA and protein expression of BMP6 / BMP7 in bone of glucocorticoid-induced osteoporosis rats, to reveal the pathogenesis of glucocorticoid osteoporosis and to elucidate the regulatory mechanism of nano-sized calcium and kidney medicine. Methods: The model of osteoporosis was established by injection of glucocorticoid. The experimental rats were treated with Nano-Calcium and Kidney-Reinforcing Kidney with high-dose and low-dose of Kidney-benefiting kidney essence and calcium supplement for 8 weeks. Granules, Capping Tianlian calcium as a positive control group, normal rats and model blank group as a blank control group. The mRNA and protein expression of BMP6 / BMP7 in osteoporosis rats were detected by PCR and Western blot. Results: BMP6 and BMP7 were expressed in the normal rat bone at gene and protein levels, while BMP6 / BMP7 mRNA and protein expression in the bone of model rats were significantly decreased. BMP-7 / BMP7 mRNA and protein expression were significantly increased in high and low dose groups, GTI group and Gushukang group after 8 weeks of treatment compared with those in model group. CONCLUSION: Nano-Calcium Bushen Chinese medicine can prevent and treat bone formation and promote glucocorticoid-induced osteoporosis by regulating BMP6 / BMP7 mRNA and protein expression in rat bone.
