目的 探讨错配修复基因hMLH1在原发性肝细胞癌(HCC)组织中的表达及其意义。方法 采用免疫组织化学和逆转录 聚合酶链反应(RT PCR)检测48例肝癌组织、2 3例癌旁组织和2 5例正常肝组织中hMLH1蛋白和mRNA的表达情况,同时采用原位末端标记法测定癌组织的细胞凋亡指数。结果 48例肝癌组织、2 3例癌旁组织和2 5例正常肝组织的hMLH1mRNA表达分别为62 .5 % (3 0 /4 8)、10 0 .0 % (2 3 /2 3 )和10 0 .0 % (2 5 /2 5 ) ,蛋白表达分别为60 .41% (2 9/4 8)、91.3 0 % (2 1/2 3 )和10 0 .0 0 % (2 5 /2 5 )。hMLH 1表达与肝癌组织的临床分期有明显相关,hMLH1表达阳性的肝癌组织的细胞凋亡指数升高为(3 .5 62±0 .2 3 3 ) %。结论 hMLH1在肝癌组织中低表达,提示错配修复基因hMLH1功能缺陷在肝癌的发生、发展过程中起重要作用
Objective To investigate the expression of mismatch repair gene hMLH1 in primary hepatocellular carcinoma (HCC) and its significance. Methods Immunohistochemistry and reverse transcription polymerase chain reaction (RT PCR) were used to detect the expression of hMLH1 protein and mRNA in 48 cases of HCC, 23 cases of paracancerous tissues and 25 cases of normal liver tissues. The expression of hMLH1 protein and mRNA was detected by in situ end-labeling Method to determine the apoptosis index of cancer tissue. Results The expression of hMLH1 mRNA in 48 cases of HCC, 23 cases of paracancerous tissues and 25 cases of normal liver tissues were 62.5% (30/40), 100.0% (23/3) and 10 The protein expression was 60.41% (2 9/4 8), 91.3 0% (2 1/2 3) and 100.0% (2 5/2 5). The expression of hMLH 1 was significantly correlated with the clinical stage of HCC tissues. The apoptosis index of hMLH1 positive HCC tissues was (3.62 ± 0.23)%. Conclusion The low expression of hMLH1 in HCC tissues suggests that the dysfunction of mismatch repair gene hMLH1 plays an important role in the development and progression of HCC
